| 3 | Ginsenosides stimulated the proliferation of mouse spermatogonia involving activation of protein kinase C显示文摘The effect of ginsenosides on proliferation of type A spermatogonia was investigated in 7-day-old mice. Spermatogonia were characterized by c-kit expression and cell proliferation was assessed by immunocytochemical demonstration of proliferating cell nuclear antigen (PCNA). After 72-h culture, Sertoli cells formed a confluent monolayer to which numerous spermatogonial colonies attached. Spermatogonia were positive for c-kit staining and showed high proliferating activity by PCNA expression. Ginsenosides (1.0~10 μg/ml) significantly stimulated proliferation of spermatogonia. Activation of protein kinase C (PKC) elicited proliferation of spermatogonia at 10-8 to 10-7 mol/L and the PKC inhibitor H7 inhibited this effect. Likewise, ginsenosides-stimulated spermatogonial proliferation was suppressed by combined treatment of H7. | 4 | 白术对猪O型口蹄疫疫苗免疫效果及抗氧化性的影响显示文摘为提高猪O型口蹄疫疫苗的免疫效果,将试验猪分为5组,在免疫口蹄疫疫苗前分别饲喂基础日粮、添加了0.5%粉碎白术、1.0%粉碎白术、0.5%煎煮白术、1.0%煎煮白术的试验日粮,然后对各组猪进行口蹄疫的常规免疫,首免前和二免后3周后分别采血测定口蹄疫抗体水平.结果表明,首免前各试验组猪与对照组相比,除饲喂1.0%粉碎白术组外,口蹄疫抗体水平与对照组相比差异均不显著(P>0.05),且低于试剂盒0.4的阳性标准,二免后3周的测定结果表明,0.5%水平的粉碎白术和0.5%煎煮白术均较对照组显著提高了口蹄疫的抗体水平(P<0.05),试验中未见明显不良反应,对试验猪体增重的影响也不大;此外,首免前饲喂1.0%粉碎白术组的抗体水平均值为0.409,高于试剂盒0.4的阳性标准,且显著高于对照组(P<0.05),二免后3周该组的均值为0.384,低于试剂盒0.4的阳性标准,与对照组差异不显著(P>0.05).总之,0.5%的煎煮或粉碎白术添加到基础日粮饲喂10 d,可显著提高O型口蹄疫疫苗二免后3周的口蹄疫抗体水平,生产中母源抗体对口蹄疫疫苗免疫失败的影响也值得重视. | 李瑞丽 严寒 郑小明 万明春 | 2019 | 猪业科学2019,36,8: | 1 |

| 7 | Investigation on Prescription Screening of a Polyherbal Preparation Based on Splenocyte Proliferation Activity and its Preparation Method显示文摘Objective: The present study aims to optimize the extraction conditions of polysaccharides and flavonoids from a polyherbal preparation consisting of three kinds of Chinese medicinal herbs, Codonopsis pilosula, Crataegus pinnatifida, and Lycium barbarum, and evaluation of its immunomodulatory activity in immunosuppressed mice. Materials and Methods: An orthogonal design(L_9 [3]~4) was constructed to achieve the optimal extraction conditions. The immunomodulatory action of the polyherbal preparation was studied at three doses of 10, 20, and 40 mL/kg/day orally by measuring splenocyte proliferation in mice model of cyclophosphamide-induced immunosuppression. Results: The chosen parameters, including the ratio of solvent-to-raw material, duration of extraction, and extraction times, were the fundamental variables that influenced the extract yields. The highest yield of total polysaccharides content was 54.3 mg/mL when the ratio of solvent-to-raw material,duration of extraction, and number of extractions were 12:1, 1.5 h, and 3, respectively. The maximum extraction yield of the flavonoids was 3.5 mg/mL when the ratio of solvent-to-raw material was 12:1, the extraction time was 2 h, and the number of extractions was 3. The prescription screening showed that the impact of the polyherbal preparation on the splenocyte proliferation capacity was more pronounced than its disassembled components. Oral administration of the polyherbal preparation could significantly increase the concanavalin A-stimulated mouse spleen cells proliferation in a dose-dependent manner.
