| 1 | TNF-α在D-GalN/LPS诱导急性肝衰竭中的作用研究显示文摘急性肝衰竭是严重威胁人类健康的疾病之一,其特征表现为机体短期内出现大面积炎症反应和大量肝细胞死亡。D-氨基半乳糖联合脂多糖常被用于构建急性肝衰竭动物模型。大量的研究证实,肿瘤坏死因子-α与急性肝衰竭密切相关。本文拟就肿瘤坏死因子-α在D-氨基半乳糖/脂多糖诱导肝损伤发生、发展中所起的作用进行综述,重点讨论了肿瘤坏死因子-α的表达机制、诱导肝细胞凋亡途径及药物干预作用,以期对急性肝衰竭的防治提供新的靶点和方向。 | 郑侠 范启林 许露露 张淼 张玉彬 | 2017 | 药学研究2017,36,7: | 7 |
| 2 | 番茄红素抑制内质网应激从而减轻CCl_4诱导的小鼠急性肝损伤显示文摘目的:探究番茄红素对四氯化碳(carbon tetrachloride,CCl_4)诱导的急性肝损伤小鼠内质网应激(endoplasmic reticulum stress,ERS)的作用及作用机制。方法:灌胃给予番茄红素7 d后,用CCl_4复制小鼠急性肝损伤模型,试剂盒检测肝组织乳酸脱氢酶(lactic dehydrogenase,LDH)活性;TUNEL检测细胞凋亡;Western blot检测ERS相关蛋白及PI3K/Akt通路标记蛋白的表达。结果:番茄红素能降低肝脏LDH(P<0.01)活性,同时还可明显抑制细胞凋亡(P<0.05);此外。番茄红素能明显抑制ERS相关蛋白葡萄糖调节蛋白(glucose regulated protein 78kda,GRP78)、激活转录因子(activating trnsfection factor-6,ATF-6)、p-肌醇酶-1α(inositol requiring enzyme,p-IRE-1α)、p-真核起始因子-2α(eukaryotic initiation factor-2α,p-e IF-2α)和caspase-12的表达(P<0.05),升高p-Akt/Akt的比值(P<0.05)。结论:番茄红素可通过抑制内质网氧化应激减轻CCl4诱导的肝损伤,其机制可能与促进PI3K/Akt通路激活有关。 | 廖笑玲 吴亮 林宗泽 叶剑 | 2018 | 中国临床药理学与治疗学2018,23,7: | 5 |
| 3 | Effect of Chinese herbal compound on liver fibrosis in rabbits with schistosomiasis by B-ultrasound显示文摘Objective: To explore the value of B-ultrasound on the evaluation of the effects of traditional Chinese medicine compound of Radix astragali , Salvia miltiorrhiza and Angelica sinensis , and TCM + praziquantel on liver fibrosis in rabbits with schistosomiasis. Methods: The hepatic fibrosis model in rabbits with schistosomiasis was established. The experimental animals (24 rabbits) were randomly divided into four groups (group A, B, C and D, n =6). Group A (control group) was only treated by praziquantel; Group B was treated by mixture of Radix astragali and Salvia miltiorrhiza + praziquantel; Group C was treated by mixture of Radix astragali and Angelica sinensis + praziquantel; Group D was treated by mixture of Radix astragali, Salvia miltiorrhiza and Angelica sinensis + praziquantel. Then B-ultrasonogram was used to evaluate the effects. Results: Each group showed certain curative effect on liver fibrosis in rabbits with schistosomiasis. The efficacy of group B, C and D was better than group A, and that of group D was the best. The differences in long diameter, thickness diameter, transverse diameter and portal vein inner diameter of liver before and after treatment were statistically significant ( P <0.05). The liver function indexes and liver fibrosis indexes were significantly improved after treatment ( P <0.05). | 4 | 芦丁对全氟辛酸所致的小鼠脂代谢紊乱及肾脏损伤的改善作用显示文摘目的 探讨芦丁对全氟辛酸(PFOA)所致小鼠脂代谢紊乱及肾脏损伤的改善作用。方法 将15只雄性小鼠随机分为对照组、PFOA组、PFOA+芦丁组,每组5只。PFOA组给予灌胃PFOA溶液20 mg/(kg·d),PFOA+芦丁组给予灌胃PFOA溶液20 mg/(kg·d)及芦丁溶液20 mg/(kg·d),对照组给予灌胃ddH_(2)O,连续干预14 d。每天记录小鼠体重,干预14 d后处死小鼠,采用HE染色及基底膜过碘酸六胺银(PASM)染色观察小鼠肾脏组织病理变化,并检测小鼠血清三酰甘油、总胆固醇、HDL-C、LDL-C、尿素、肌酐水平及乳酸脱氢酶(LDH)活性。结果 (1)从干预第6天开始,PFOA组和PFOA+芦丁组小鼠的体重均低于对照组(均P<0.05),而PFOA组与PFOA+芦丁组比较差异无统计学意义(均P>0.05)。(2)HE染色和PASM染色结果显示,与对照组相比,PFOA组小鼠肾脏出现明显的结构性损伤,肾小球体积增大,系膜细胞和内皮细胞增生,肾小管上皮细胞水肿、脱落及轻度空泡样变性,肾小球系膜增厚,肾小管基底膜增厚、皱缩,近端小管刷状缘消失;而与PFOA组相比,PFOA+芦丁组小鼠的肾脏组织损伤明显减轻。(3)PFOA组的血清总胆固醇、LDL-C、尿素水平均高于对照组,血清肌酐水平低于对照组;PFOA+芦丁组的血清总胆固醇、LDL-C、尿素水平均低于PFOA组,血清肌酐水平高于PFOA组(均P<0.05);3组小鼠的血清三酰甘油、HDL-C水平及LDH活性比较,差异均无统计学意义(均P>0.05)。结论 芦丁可改善PFOA造成的小鼠肾脏病理学变化及肾功能损伤,对小鼠肾脏具有一定保护作用,并有助于调节其脂质代谢。 | 王广胤 孙伟强 马新壮 王力 | 2022 | 广西医学2022,44,17: | 1 |
