1Oncofetal antigen glypican-3 as a promising early diagnostic marker for hepatocellular carcinoma显示文摘BACKGROUND:Hepatocellular carcinoma (HCC) is characterized by a multi-cause,multi-stage and multi-focus process of tumor progression.Its prognosis is poor and early diagnosis is of utmost importance.This study was undertaken to investigate the dynamic expression of oncofetal antigen glypican-3 (GPC-3) and GPC-3 mRNA in hepatocarcinogenesis and to explore their early diagnostic value for HCC.METHODS:A hepatoma model was induced in male Sprague-Dawley rats with 0.05% 2-fluorenylacetamide and confirmed by hematoxylin and eosin staining and gamma-glutamyltransferase (GGT) expression.Total RNA was purified and transcribed into cDNA by reverse transcription.Fragments of the GPC-3 gene were amplified by nested RT-PCR,and confirmed by sequencing.GPC-3 was analyzed by immunohistochemistry,Western blotting or ELISA.RESULTS:Positive GPC-3 expression showed as brown granule-like staining localized in the cytoplasm.Histological examination of hepatocytes revealed three morphological stages of granule-like degeneration,atypical hyperplasia (precancerous),and cancer formation,with a progressive increase of liver total RNA and GGT expression.The incidence of liver GPC-3 mRNA and GPC-3,and serum GPC-3 was 100%,100% and 77.8% in the HCC group,100%,100%,and 66.7% in the precancerous group,83.3%,83.3%,and 38.9% in the degeneration group,and no expression in the liver or blood of the control group,respectively.There was a positive correlation between liver GPC-3 mRNA and total RNA level (r=0.475,P<0.05) or liver GPC-3 (r=1.0,P<0.001) or serum GPC-3 (r= 0.994,P<0.001).CONCLUSION:Abnormal oncofetal antigen GPC-3 and GPC-3 mRNA expression in hepatocarcinogenesis may be promising molecular markers for early diagnosis of HCC.Research Center of Clinical Medicine,Affiliated Hospital of Nantong University,Nantong 226001,China (Yao M,Yao DF,Qiu LW,Wu W,Sai WL,Yang JL and Zhang HJ) Department of Oncology,Yancheng First People’s Hospital,Yancheng 224001,China (Bian YZ) Department of Oncology,Second Affiliated Hospital,Nanjing Medical University,Nanjing 210011,China (Zhang CG) 2011Hepatobiliary & Pancreatic Diseases International2011,10,3:51
2Quantitative analysis of hepatic hypoxia-inducible factor-1α and its abnormal gene expression during the formation of hepatocellular carcinoma显示文摘BACKGROUND:Hepatic hypoxia-inducible factor-1(HIF-1) is activated in the progression of hepatocellular carcinoma (HCC).This study aimed to investigate the dynamic alterations of HIF-1αand its gene expression so as to explore the relationship between HIF-1αexpression and hepatocarcinogenesis at the early stage of HCC. METHODS:A hepatoma model was made with 2-fluorenyl- acetamide(2-FAA)in male Sprague-Dawley rats.Morphological changes of rat hepatocytes were assessed pathologically (HE staining).The dynamic expression of hepatic and circulating HIF-1αwas quantitatively analyzed by ELISA. The gene fragments of hepatic HIF-1αmRNA were amplified by RT-PCR and confirmed by sequencing.The cellular distribution of hepatic HIF-1αexpression was confirmed by immunohistochemistry. RESULTS:Histological examination confirmed granulelike degeneration to atypical hyperplasia and HCC development in rat hepatocytes and progressive increases in the levels of hepatic and circulating HIF-1αand its gene expression during the course.The levels of HIF-1α expression in the liver and blood of rats with hepatoma were significantly higher than those in normal ratsand those with degeneration.Immunohistochemical analysis confirmed the positive expression and hepatocyte distribution of HIF-1αin the development of rat hepatoma. A positive relationship was found between HIF-1α expression in the liver and blood(P<0.01). 3Structure and function of aggrecan显示文摘Aggrecan is the major proteoglycan in the articular cartilage. This molecule is important in the proper functioning of articular cartilage because it provides a hydrated gel structure (via its interaction with hyaluronan and link protein) that endows the cartilage with load-bearing properties. It is also crucial in chondroskeletal morphogenesis during development. Aggrecan is a multimodular molecule expressed by chondrocytes. Its core protein is composed of three globular domains (G1, G2, and G3) and a large extended region (CS) between G2 and G3 for glycosaminoglycan chain attachment. G1 comprises the amino terminus of the core protein. This domain has the same structural motif as link protein. Functionally, the G1 domain interacts with hyaluronan acid and link protein, forming stable ternary complexes in the extracellular matrix. G2 is homologous to the tandem repeats of G1 and of link protein and is involved in product processing. G3 makes up the carboxyl terminus of the core protein. It enhances glycosaminoglycan modification and product secretion. 6Priorities for research and control of cestode zoonoses in Asia显示文摘Globally,cestode zoonoses cause serious public health problems,particularly in Asia.Among all neglected zoonotic diseases,cestode zoonoses account for over 75%of global disability adjusted life years(DALYs)lost.An international symposium on cestode zoonoses research and control was held in Shanghai,China between 28th and 30th October 2012 in order to establish joint efforts to study and research effective approaches to control these zoonoses.It brought together 96 scientists from the Asian region and beyond to exchange ideas,report on progress,make a gap analysis,and distill prioritizing settings with a focus on the Asian region.Key objectives of this international symposium were to agree on solutions to accelerate progress towards decreasing transmission,and human mortality and morbidity caused by the three major cestode zoonoses(cystic echinococcosis,alveolar echinococcosis,and cysticercosis);to critically assess the potential to control these diseases;to establish a research and validation agenda on existing and new approaches;and to report on novel tools for the study and control of cestode zoonoses.Ning Xiao Jia-Wen Yao Wei Ding Patrick Giraudoux Philip S Craig Akira Ito 2013Infectious Diseases of Poverty2013,2,1:6
7Orthogonal design for optimization of pigment extraction from surface sediments of the Changjiang River Estuary显示文摘Using a suitable solvent for extracting pigments from sediment for high performance liquid chromatography(HPLC) analysis is critical for obtaining qualitative and quantitative estimates of phytoplanktonic and benthic algal biomass,as well as community composition. Five methodological factors(sample dehydration,extraction solvent,extraction duration,number of extractions,and ratio of solvent volume:sample weight) were studied using an L 9(3 4 ) orthogonal design in a sedimentary pigment extraction experiment on samples collected from the Changjiang large-river delta-front estuary(LDE),using HPLC analysis. The results show that the optimal extraction method for sedimentary pigments should include freeze-drying samples prior to extraction. The effects of different factors on sedimentary pigment extraction were separated by the L 9(3 4 ) orthogonal design experiments and showed that the extraction solvent was the most important,with extraction duration the second most important,and numbers of extraction and ratio of solvent volume:sample weight was the least important. The mixed solvent treatment comprised of acetone,methanol and water(80:15:5,by volume) was best for polar pigment extraction,with 100% acetone better for apolar pigments. For most pigments employed in this study(i.e.,peridinin,fucoxanthin,alloxanthin,diatoxanthin,zeaxanthin,pheophytin-a and β-carotene) ,3 h was found to be enough time for extraction from these deltaic sediments. However,for chlorophyll-a,the most important pigment used for estimating algal biomass,12 h was needed. A small amount of solvent(3 ml) with duplicate extractions obtained the greatest amount and diversity of pigments. Unfortunately,no extraction method was found to be suitable for all pigments in sediments. 8Molecular and Epidemiological Characterization of Infant Botulism in Beijing, China显示文摘Laboratory-based pathogen isolation, identification, and toxicity determination were performed on samples from a suspected case of infant botulism. Mice injected with cultures generated from the enema sample and ingested Powered infant formula(PIF) presented typical signs of botulism. Antitoxins to polyvalent botulinum neurotoxins(BoNTs) and monovalent BoNT type B antitoxin had protective effects. Clostridium botulinum isolated from the enema and residual PIF samples were positive for type B toxin. Pulsed-field gel electrophoresis(PFGE) revealed that the two strains of C. botulinum isolated from the two samples produced indistinguishable pulsotypes.
