1Relative Contributions of Boundary-Layer Meteorological Factors to the Explosive Growth of PM2.5 during the Red-Alert Heavy Pollution Episodes in Beijing in December 2016显示文摘Based on observations of urban mass concentration of fine particulate matter smaller than 2.5 μm in diameter(PM_(2.5)), ground meteorological data, vertical measurements of winds, temperature, and relative humidity(RH), and ECMWF reanalysis data, the major changes in the vertical structures of meteorological factors in the boundary layer(BL) during the heavy aerosol pollution episodes(HPEs) that occurred in winter 2016 in the urban Beijing area were analyzed. The HPEs are divided into two stages: the transport of pollutants under prevailing southerly winds, known as the transport stage(TS), and the PM_(2.5) explosive growth and pollution accumulation period characterized by a temperature inversion with low winds and high RH in the lower BL, known as the cumulative stage(CS). During the TS, a surface high lies south of Beijing, and pollutants are transported northwards. During the CS, a stable BL forms and is characterized by weak winds, temperature inversion, and moisture accumulation. Stable atmospheric stratification featured with light/calm winds and accumulated moisture(RH > 80%) below 250 m at the beginning of the CS is closely associated with the inversion, which is strengthened by the considerable decrease in near-surface air temperature due to the interaction between aerosols and radiation after the aerosol pollution occurs. A significant increase in the PLAM(Parameter Linking Aerosol Pollution and Meteorological Elements) index is found, which is linearly related to PM mass change. During the first 10 h of the CS, the more stable BL contributes approximately 84% of the explosive growth of PM_(2.5) mass. Additional accumulated near-surface moisture caused by the ground temperature decrease, weak turbulent diffusion, low BL height, and inhibited vertical mixing of water vapor is conducive to the secondary aerosol formation through chemical reactions, including liquid phase and heterogeneous reactions, which further increases the PM_(2.5) concentration levels. 2Review grain yield and nitrogen use efficiency in rice production regions in China显示文摘As one of the staple food crops, rice(Oryza sativa L.) is widely cultivated across China, which plays a critical role in guaranteeing national food security. Most previous studies on grain yield or/and nitrogen use efficiency(NUE) of rice in China often involved site-specific field experiments, or small regions with insufficient data, which limited the representation for the current rice production regions. In this study, a database covering a wide range of climate conditions, soil types and field managements across China, was developed to estimate rice grain yield and NUE in various rice production regions in China and to evaluate the relationships between N rates and grain yield, NUE. According to the database for rice, the values of grain yield, plant N accumulation, N harvest index(HIN), indigenous N supply(INS), internal N efficiency(IE_N), reciprocal internal N efficiency(RIE_N), agronomic N use efficiency(AE_N), partial N factor productivity(PEPN), physiological N efficiency(PE_N), and recover efficiency of applied N(RE_N) averaged 7.69 t ha^(–1), 152 kg ha^(–1), 0.64 kg kg^(–1), 94.1 kg kg^(–1), 53.9 kg kg^(–1), 1.98 kg kg^(–1), 12.6 kg kg^(–1), 48.6 kg kg^(–1), 33.8 kg kg^(–1), and 39.3%, respectively. However, the corresponding values all varied tremendously with large variation. Rice planting regions and N rates had significant influence on grain yield, N uptake and NUE values. Considering all observations, N rates of 200 to 250 kg ha^(–1) commonly achieved higher rice grain yield compared to less than 200 kg N ha^(–1) and more than 250 kg N ha^(–1) at most rice planting regions. At N rates of 200 to 250 kg ha^(–1), significant positive linear relationships were observed between rice grain yield and AE_N, PE_N, RE_N, IE_N, and PFPN, and 46.49, 24.64, 7.94, 17.84, and 88.24% of the variation in AE_N, PE_N, RE_N, IE_N, and PFPN could be explained by grain yield, respectively. 3Rainfall and inflow effects on soil erosion for hillslopes dominated by sheet erosion or rill erosion in the Chinese Mollisol region显示文摘Erosion agents and patterns profoundly affect hillslope soil loss characteristics. However, few attempts have been made to analyze the effects of rainfall and inflow on soil erosion for hillslopes dominated by sheet erosion or rill erosion in the Chinese Mollisol region. The objective of this study was to discuss the erosive agent(rainfall or inflow), hillslope erosion pattern(sheet erosion or rill erosion) and slope gradient effects on runoff and soil losses. Two soil pans(2.0 m long, 0.5 m wide and 0.5 m deep) with 5° and 10° slopes were subjected to rainfall(0 and 70 mm h–1) and inflow(0 and 70 mm h–1) experiments. Three experimental combinations of rainfall intensity(RI) and inflow rate(IR) were tested using the same water supply of 70 mm by controllingthe run time. A flat soil surface and a soil bed with a straight initial rill were prepared manually, and represented hillslopes dominated by sheet erosion and rill erosion, respectively. The results showed that soil losses had greater differences among treatments than total runoff. Soil losses decreased in the order of RI70+IR70 > RI70+IR0 > RI0+IR70. Additionally, soil losses for hillslopes dominated by rill erosion were 1.7-2.2 times greater at 5° and 2.5-6.9 times greater at 10° than those for hillslopes dominated by sheet erosion. The loss of <0.25 mm soil particles and aggregates varying from 47.72%-99.60% of the total soil loss played a dominant role in the sediment. Compared with sheet erosion hillslopes, rill erosion hillslopes selectively transported more microaggregates under a relatively stable rill development stage, but rills transported increasinglymore macroaggregates under an active rill development stage. 4Endothelial cell-specific anticoagulation reduces inflammation in a mouse model of acute lung injury显示文摘Tissue factor (TF)-dependent coagulation contributes to lung inflammation and the pathogenesis of acute lung injury (ALI). In this study, we explored the roles of targeted endothelial anticoagulation in ALI using two strains of transgenic mice expressing either a membrane-tethered human tissue factor pathway inhibitor (hTFPI) or hirudin fusion protein on CD31^+ cells, including vascular endothelial cells (ECs). ALI was induced by intratracheal injection of LPS, and after 24 h the expression of TF and protease-activated receptors (PARs) on EC in lungs were assessed, alongside the extent of inflammation and injury. The expression of TF and PARs on the EC in lungs was upregulated after ALI. In the two strains of transgenic mice, expression of either of hTFPI or hirudin by EC was associated with significant reduction of inflammation, as assessed by the extent of leukocyte infiltration or the levels of proinflammatory cytokines, and promoted survival after LPS-induced ALI. The beneficial outcomes were associated with inhibition of the expression of chemokine CCL2 in lung tissues. The protection observed in the CD31-TFPI-transgenic strain was abolished by injection of an anti-hTFPI antibody, but not by prior engraftment of the transgenic strains with WT bone marrow, confirming that the changes observed were a specific transgenic expression of anticoagulants by EC. These results demonstrate that the inflammation in ALI is TF and thrombin dependent, and that expression of anticoagulants by EC significantly inhibits the development of ALI via repression of leukocyte infiltration, most likely via inhibition of chemokine gradients. 5Aerosol Hygroscopicity during the Haze Red-Alert Period in December 2016 at a Rural Site of the North China Plain显示文摘A humidification system was deployed to measure aerosol hygroscopicity at a rural site of the North China Plain during the haze red-alert period 17–22 December 2016. The aerosol scattering coefficients under dry [relative humidity(RH) < 30%] and wet(RH in the range of 40%–85%) conditions were simultaneously measured at wavelengths of450, 550, and 700 nm. It is found that the aerosol scattering coefficient and backscattering coefficient increased by only 29% and 10%, respectively when RH went up from 40% to 80%, while the hemispheric backscatter fraction went down by 14%, implying that the aerosol hygroscopicity represented by the aerosol scattering enhancement factor f(RH) is relatively low and RH exerted little effects on the aerosol light scattering in this case. The scattering enhancement factors do not show significant differences at the three wavelengths, only with an approximate 2% variation, suggesting that the aerosol hygroscopicity is independent of the wavelength. Aerosol hygroscopicity is highly dependent on the aerosol chemical composition. When there is a large mass fraction of inorganics and a small mass fraction of organic matter, f(RH) reaches a high value. 6Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5
7Chemical Components, Variation, and Source Identification of PM1 during the Heavy Air Pollution Episodes in Beijing in December 2016显示文摘Air pollution is a current global concern. The heavy air pollution episodes(HPEs) in Beijing in December 2016 severely influenced visibility and public health. This study aims to survey the chemical compositions, sources, and formation processes of the HPEs. An aerodyne quadruple aerosol mass spectrometer(Q-AMS) was utilized to measure the non-refractory PM1(NR-PM1) mass concentration and size distributions of the main chemical components including organics, sulfate, nitrate, ammonium, and chloride in situ during 15–23 December 2016. The NR-PM1 mass concentration was found to increase from 6 to 188 μg m–3 within 5 days. During the most serious polluted episode, the PM1 mass concentration was about 2.6 times that during the first pollution stage and even 40 times that of the clean days. The formation rates of PM2.5 in the five pollution stages were 26, 22, 22, 32, and 67 μg m^(–3) h–1, respectively. Organics and nitrate occupied the largest proportion in the polluted episodes, whereas organics and sulfate dominated the submicron aerosol during the clean days. The size distribution of organics is always broader than those of other species, especially in the clean episodes. The peak sizes of the interested species grew gradually during different HPEs. Aqueous reaction might be important in forming sulfate and chloride, and nitrate was formed via oxidization and condensation processes. PMF(positive matrix factorization) analysis on AMS mass spectra was employed to separate the organics into different subtypes. Two types of secondary organic aerosol with different degrees of oxidation consisted of 43% of total organics. 8Nutrient uptake requirements with increasing grain yield for rice in China显示文摘Improved estimates of nutrient requirements for rice(Oryza sativa L.)in China are essential to optimize fertilization regulation for increasing grain yields and reducing the potential of environmental negative influences,especially under high-yielding intensive systems.A database involving rice grain yields,nutrient concentrations and accumulations collected from on-field station experiments in the literatures published from 2000 to 2013 in China was developed to understand the relationships between grain yields and plant nutrient uptakes,and to quantify nutrient requirements for different yield levels.Considering all data sets,rice grain yield ranged from 1.4 to 15.2t ha^(-1)with the mean value of 7.84t ha^(-1),and ca.10.4%of yield observations were higher than the yield barrier level of 10 t ha^(-1).N requirement to produce one ton grain was 21.10 kg for the yield range<4.0t ha^(-1)with a high variation of 45.8%.Except of the yield range<4.0t ha^(-1),the values of N requirement,firstly increased from 18.78 kg for yield range 4.0-5.5 t ha^(-1)to 20.62 kg for yield range 7.0-8.5 t ha^(-1),then decreased slightly to 19.67 and19.17 kg for the yield range 8.5-10 and>10 t ha^(-1),respectively.Phosphorus(P)and potassium(K)requirements showed increasing trends,from 3.51 and 19.87 kg per t grain for<4.0 t ha^(-1)yield range to 4.10 and 21.70 kg for>10.0t ha^(-1)range.In conclusion,nutrient requirement varied with increasement of grain yield,and N,P and K presented various response trends,increasing,declining or stagnating,which would be of great benefit for improving fertilizer strategies.CHE Sheng-guo ZHAO Bing-qiang LI Yan-ting YUAN Liang LIN Zhi-an HU Shu-wen SHEN Bing 2016Journal of Integrative Agriculture2016,15,4:4

10Transfer of β-hydroxy-β-methylbutyrate from sows to their offspring and its impact on muscle fiber type transformation and performance in pigs显示文摘Background: Previous studies suggested that supplementation of lactating sows with β-hydroxy-β-methylbutyrate(HMB) could improve the performance of weaning pigs, but there were little information in the muscle fiber type transformation of the offspring and the subsequent performance in pigs from weaning through finishing in response to maternal HMB consumption. The purpose of this study was to determine the effect of supplementing lactating sows with HMB on skeletal muscle fiber type transformation and growth of the offspring during d 28 and180 after birth. A total of 20 sows according to their body weight were divided into the control(CON, n = 10) or HMB groups(HMB, n = 10). Sows in the HMB group were supplemented with β-hydroxy-β-methylbutyrate calcium(HMB-Ca) 2 g/kg feed during d 1 to 27 of lactation. After weaning, 48 mixed sex piglets were blocked by sow treatment and fed standard diets for post-weaning, growing, finishing periods. Growth performance was recorded during d 28 to 180 after birth. Pigs were slaughtered on d 28(n = 6/treatment) and 180(n = 6/treatment) postnatal and the longissimus dorsi(LD) was collected, respectively.Results: The HMB-fed sows during lactation showed increased HMB concentration(P < 0.05) in milk and LD of weaning piglets(P < 0.05). 12Robust prediction of hourly PM_(2.5) from meteorological data using LightGBM显示文摘Retrieving historical fine particulate matter(PM_(2.5))data is key for evaluating the long-term impacts of PM_(2.5) on the environment,human health and climate change.Satellite-based aerosol optical depth has been used to estimate PM_(2.5),but estimations have largely been undermined by massive missing values,low sampling frequency and weak predictive capability.Here,using a novel feature engineering approach to incorporate spatial effects from meteorological data,we developed a robust Light GBM model that predicts PM_(2.5) at an unprecedented predictive capacity on hourly(R^(2)=0.75),daily(R^(2)=0.84),monthly(R^(2)=0.88)and annual(R^(2)=0.87)timescales.By taking advantage of spatial features,our model can also construct hourly gridded networks of PM_(2.5).This capability would be further enhanced if meteorological observations from regional stations were incorporated.Our results show that this model has great potential in reconstructing historical PM_(2.5) datasets and real-time gridded networks at high spatial-temporal resolutions.The resulting datasets can be assimilated into models to produce long-term re-analysis that incorporates interactions between aerosols and physical processes.Junting Zhong Xiaoye Zhang Ke Gui Yaqiang Wang Huizheng Che Xiaojing Shen Lei Zhang Yangmei Zhang Junying Sun Wenjie Zhang 2021National Science Review2021,8,10:2
13Comparison of Submicron Particles at a Rural and an Urban Site in the North China Plain during the December 2016 Heavy Pollution Episodes显示文摘An extensive field experiment for measurement of physical and chemical properties of aerosols was conducted at an urban site in the Chinese Academy of Meteorological Sciences(CAMS) in Beijing and at a rural site in Gucheng(GC), Hebei Province in December 2016. This paper compares the number size distribution of submicron particle matter(PM1, diameter < 1 μm) between the two sites. The results show that the mean PM1 number concentration at GC was twice that at CAMS, and the mass concentration was three times the amount at CAMS. It is found that the accumulation mode(100–850 nm) particles constituted the largest fraction of PM1 at GC, which was significantly correlated with the local coal combustion, as confirmed by a significant relationship between the accumulation mode and the absorption coefficient of soot particles. The high PM1 concentration at GC prevented the occurrence of new particle formation(NPF) events, while eight such events were observed at CAMS. During the NPF events, the mass fraction of sulfate increased significantly, indicating that sulfate played an important role in NPF. The contribution of regional transport to PM1 mass concentration was approximately 50% at both sites, same as that of the local emission. 14miR-150 regulates glucose utilization through targeting GLUT4 in insulin-resistant cardiomyocytes显示文摘MicroRNAs(miRNAs)play an important role in cardiac function and metabolism.However,whether they regulate insulin resistance(IR)of cardiomyocytes remains unclear.The aim of the present study was to shed light on this issue with a focus on miR-150.We found here that miR-150 level was elevated in myocardium of type 2 diabetes mellitus(T2DM)rat model and in insulin-resistant cardiomyocytes induced by high glucose(25 mM)and high insulin(1μM).Deregulation of miR-150 downregulated the protein and mRNA levels of glucose transporter 4(GLUT4)as assessed by western blot,real-time polymerase chain reaction(qPCR),and immunofluorescence assays.Overexpression of miR-150 inhibited glucose utilization in cardiomyocytes as detected by 2-deoxyglucose transport and glucose consumption assays.In contrast,knockdown of miR-150 significantly increased glucose uptake in cardiomyocytes.Moreover,GLUT4 translocation was increased after transfection of miR-150 inhibitor(AMO-150).Collectively,miR-150 reduced glucose utilization by directly decreasing the expression and translocation of GLUT4 in the cardiomyocytes with IR and therefore might be a new therapeutic target for metabolic diseases such as T2DM.Jin Ju Dan Xiao Nannan Shen Tong Zhou Hui Che Xia Li Shuqian Zhang Justine Nyakango Mokembo Nabanit Kumar Jha Seth Mikaye Monayo Zhiguo Wang Yong Zhang 2020Acta Biochimica et Biophysica Sinica2020,52,10:2
