1Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment.Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen 2018Cancer Communications2018,38,1:20

3Model of nonlinear coupled thermo-hydro-elastodynamics response for a saturated poroelastic medium显示文摘Based on the Biot’s wave equation and theory of thermodynamic, Darcy law of fluid and the modified Fourier law of heat conduction, a nonlinear fully coupled thermo-hydro-elastodynamic response model (THMD) for saturated porous medium is derived. The compressibility of the medium, the influence of fluid flux on the heat flux, and the influence of change of temperature on the fluid flux are considered in this model. With some simplification, the coupled nonlinear thermo-hydro-elastodynamic response model can be reduced to the thermo-elastodynamic (TMD) model based on the traditional Fourier law and, further more, to the Biot’s wave equation without considering the heat phase. 4Internal tide characteristics over northern South China Sea continental slope显示文摘21-day in-situ ADCP current and CTD data from an anchored observational station by R/V Shiyan 3 are used to investigate the characteristics of internal tide in the northern South China Sea continental slope. The results show that tide and internal tide are both mixed type in the observed region and dominated by diurnals; the maximum internal tidal major axes of K1, O1 were largest and reached 11 cm/s, 10 cm/s, respectively, and their current vectors rotate anticlockwise. The 300 m-deep internal tide presents obvious quasi-diurnal oscillation and its average amplitude reaches 50 m. 5Cyclic performance and simplified pushover analyses of precast segmental concrete bridge columns with circular section显示文摘In recent years, precast segmental concrete bridge columns became prevalent because of the benefits of accelerated construction, low environmental impact, high quality and low life cycle costs. The lack of a detailed configuration and appropriate design procedure to ensure a comparable performance with monolithic construction has impeded this structural system from being widely used in areas of high seismicity. In this study, precast segmental bridge column cyclic loading tests were conducted to investigate the performance of unbonded post-tensioned segmental bridge columns. One monolithic and two precast segmental columns were tested. The precast segmental column exhibited minor damage and small residual displacement after the maximum 7% cyclic drift; energy dissipation(ED) can be enhanced byadding ED bars. The experimental results were modeled by a simplified pushover method(SPOM), as well as a fiber model(FIBM) finite element method. Forty-five cases of columns with different aspect ratios, axial load ratios and ED bar ratios were analyzed with the SPOM and FIBM, respectively. Using these parametric results, a simplified design method was suggested by regressive analysis. 6Research on Radiation Characteristics of Polymeric Plastic Fiber显示文摘The anti-radiation mechanism of polymeric plastic fiber is analyzed. The radiation characteristic of this type fiber is studied. Our experimental results indicate that under the low radiation dosage (below 1 kGy) the plastic optical fiber’s transmission rate rises instead of descends while this type of fiber is radiated by γ ray, electron beam and proton beam respectively. After a period of time, it gradually reaches a constant value. But under the high radiation dosage (above 1 kGy) the fiber’s transmission rate descends after radiation. 8Fault-tolerant control with mixed aerodynamic surfaces and RCS jets for hypersonic reentry vehicles显示文摘This paper proposes a fault-tolerant strategy for hypersonic reentry vehicles with mixed aerodynamic surfaces and reaction control systems(RCS) under external disturbances and subject to actuator faults.Aerodynamic surfaces are treated as the primary actuator in normal situations,and they are driven by a continuous quadratic programming(QP) allocator to generate torque commanded by a nonlinear adaptive feedback control law.When aerodynamic surfaces encounter faults,they may not be able to provide sufficient torque as commanded,and RCS jets are activated to augment the aerodynamic surfaces to compensate for insufficient torque.Partial loss of effectiveness and stuck faults are considered in this paper,and observers are designed to detect and identify the faults.Based on the fault identification results,an RCS control allocator using integer linear programming(ILP) techniques is designed to determine the optimal combination of activated RCS jets.By treating the RCS control allocator as a quantization element,closed-loop stability with both continuous and quantized inputs is analyzed.Simulation results verify the effectiveness of the proposed method.He Jingjing Qi Ruiyun Jiang Bin Zhai Rongyu 2017Chinese Journal of Aeronautics2017,30,2:4

10A Novel Combinational Convolutional Neural Network for Automatic Food-Ingredient Classification显示文摘With the development of deep learning and Convolutional Neural Networks(CNNs),the accuracy of automatic food recognition based on visual data have significantly improved.Some research studies have shown that the deeper the model is,the higher the accuracy is.However,very deep neural networks would be affected by the overfitting problem and also consume huge computing resources.In this paper,a new classification scheme is proposed for automatic food-ingredient recognition based on deep learning.We construct an up-to-date combinational convolutional neural network(CBNet)with a subnet merging technique.Firstly,two different neural networks are utilized for learning interested features.Then,a well-designed feature fusion component aggregates the features from subnetworks,further extracting richer and more precise features for image classification.In order to learn more complementary features,the corresponding fusion strategies are also proposed,including auxiliary classifiers and hyperparameters setting.Finally,CBNet based on the well-known VGGNet,ResNet and DenseNet is evaluated on a dataset including 41 major categories of food ingredients and 100 images for each category.Theoretical analysis and experimental results demonstrate that CBNet achieves promising accuracy for multi-class classification and improves the performance of convolutional neural networks.Lili Pan Cong Li Samira Pouyanfar Rongyu Chen Yan Zhou 2020Computers, Materials & Continua2020,,2:3

12FROST RESISTANCE OF POLYVINYL ALCOHOL FIBER REINFORCED CEMENTITIOUS COMPOSITES显示文摘This article studies the frost resistance of polyvinyl alcohol (PVA) fiber reinforced cementitious composites. The test samples contain different PVA fiber volume fractions such as 0, 0.5%, 1% and 2%. The freeze-thaw test is carried on in TDR type apparatus. 13Effects of GM-CSF, IL-3, and GM-CSG/IL-3 fusion protein on apoptosis of human myeloid leukemic cell line Tf-1 induced by irradiation显示文摘AIM: To observe the effects of three cytokines on the apoptosis of Tf-1 cells induced by γ irradiation and investigate the relationship between apoptosis and caspase-3 activity. METHODS: Different cytokines GM-CSF, IL-3 and GM-CS/IL-3 fusion protein were added into the irradiated Tf-1 cells. MTT assay, morphology, flow cytometry,and DNA fragmentation assay were used to observe the effects of cytokines on apoptosis. The caspase-3 activity was determined with a fluorocytometer. RESULTS: Irradiated Tf-1 cells showed typical morphological characteristic of apoptosis demonstrated by transmission electron microscopy and were accumulated in G0/G1 phase. In the groups treated with growth factors after irradiation, three cytokines significantly increased the viability rate, distinctly decreased the apoptosis rate and the proportion of DNA fragmentation. When Tf-1 cells were irradiated by γ irradiation, caspase-3 activity was increased at different time points. In comparison with the control group in which no growth factor was added after the cells were irradiated, the caspase-3 activity of irradiated Tf-1 cells was significantly inhibited by addition of the above cytokines. Thirty-six hours after irradiation, in the control group,GM-CSF, IL-3, GM-CSF and IL-3 in combination, and two GM-CSF/IL-3 fusion protein groups, the apoptosis ratewas 73 %, 11%, 15 %, 13 %, 12 %, and 13 %. The percent of fragmented DNA was 36 %, 19 %, 18 %, 14 %,13 %, and 14 %. The fluorescence intensity was 16923, 5529, 6581, 5322, 5426, and 5485. CONCLUSION:GM-CSF, IL-3, and GM-CSF/IL-3 fusion protein could protect Tf-1 cells from apoptosis induced by γ irradiation.After Tf-1 cells were irradiated, the caspase-3 activity was significantly increased but was dramatically decreased by the above cytokines. 17Green synthesis of gold nanoclusters using papayajuice for detection of L-lysine显示文摘Gold nanoclusters were rapid synthesized within 3 min at 120 ℃ by using papaya juice as a capping and reducing agent(P-AuNCs). The properties of the fluorescent probe were characterized by fluorescent spectroscopy, UV-vis spectroscopy, dynamic light scattering and transmission electron microscope.Based on the surface electron density increase-induced fluorescence enhancing principle, a high selective method for detection of L-lysine was developed with the as-prepared P-AuNCs coupling the fluorescence emission at 440 nm. The fluorescent probe showed high stability and good biocompatibility. Its fluorescence intensity was found to be linearly dependent on the L-lysine concentration in the range of 10.0μmol/L to 1000.0 μmol/L(R^2=0.969) with a limit of detection of 6.0μmol/L.
