Porcine Immunoglobulin Fc Fused P30/P54 Protein of African Swine Fever Virus Displaying on Surface of S. cerevisiae Elicit Strong Antibody Production in Swine显示文摘African swine fever virus(ASFV)infects domestic pigs and European wild boars with strong, hemorrhagic and high mortality. The primary cellular targets of ASFV is the porcine macrophages.Up to now,no commercial vaccine or effective treatment available to control the disease. In this study, three recombinant Saccharomyces cerevisiae(S.cerevisiae)strains expressing fused ASFV proteins-porcine Ig heavy chains were constructed and the immunogenicity of the S.cerevisiae-vectored cocktail ASFV feeding vaccine was further evaluated. To be specific, the P30-Fcγand P54-Fcαfusion proteins displaying on surface of S.cerevisiae cells were produced by fusing the Fc fragment of porcine immunoglobulin IgG1 or IgA1 with p30 or p54 gene of ASFV respectively. The recombinant P30-Fcγand P54-Fcαfusion proteins expressed by S.cerevisiae were verified by Western blotting, flow cytometry and immunofluorescence assay.Porcine immunoglobulin Fc fragment fused P30/P54 proteins elicited P30/P54-specific antibody production and induced higher mucosal immunity in swine. The absorption and phagocytosis of recombinant S.cerevisiae strains in IPEC-J2 cells or porcine alveolar macrophage(PAM)cells were significantly enhanced, too. Here,we introduce a kind of cheap and safe oral S.cerevisiae-vectored vaccine, which could activate the specific mucosal immunity for controlling ASFV infection. | Chen Chen Deping Hua Jingxuan Shi Zheng Tan Min Zhu Kun Tan Lilin Zhang Jinhai Huang | 2021 | Virologica Sinica2021,36,2: | 7

Complex interplay between formation routes and natural organic matter modification controls capabilities of C_(60)nanoparticles(nC_(60)) to accumulate organic contaminants显示文摘Accumulation of organic contaminants on fullerene nanoparticles(nC_(60)) may significantly affect the risks of C_(60) in the environment. The objective of this study was to further understand how the interplay of nC_(60) formation routes and humic acid modification affects contaminant adsorption of nC_(60).Specifically,adsorption of 1,2,4,5-tetrachlorobenzene(a model nonionic, hydrophobic organic contaminant) on nC_(60) was greatly affected by nC_(60)formation route- the formation route significantly affected the aggregation properties of nC_(60),thus affecting the available surface area and the extent of adsorption via the pore-filling mechanism.Depending on whether nC_(60) was formed via the 'top-down' route(i.e.,sonicating C_(60) powder in aqueous solution) or 'bottom-up' route(i.e.,phase transfer from an organic solvent) and the type of solvent involved(toluene versus tetrahydrofuran),modification of nC_(60) with Suwannee River humic acid(SRHA) could either enhance or inhibit the adsorption affinity of nC_(60).The net effect depended on the specific way in which SRHA interacted with C_(60) monomers and/or C_(60) aggregates of different sizes and morphology, which determined the relative importance of enhanced adsorption from SRHA modification via preventing C_(60) aggregation and inhibited adsorption through blocking available adsorption sites. The findings further demonstrate the complex mechanisms controlling interactions between nC_(60) and organic contaminants, and may have significant implications for the life-cycle analysis and risk assessment of C_(60). | Lei Hou John D.Fortner Ximeng Wang Chengdong Zhang Lilin Wang Wei Chen | 2017 | Journal of Environmental Sciences2017,29,1: | 5
Solidification Microstructure of Laser Additive Manufactured Ti—6Al—2Zr—2Sn—3Mo—1.5Cr—2Nb Titanium Alloy显示文摘Solidification microstructure of powder fed laser additive manufactured Ti—6Al—2Zr—2Sn—3Mo—1.5Cr—2Nb titanium alloy was investigated. The results showed that by deliberately increasing the powder feed rate, partially melted powders were retained at the top of the molten pool, which can promote heterogeneous nucleus. Thus, each cladding layer is composed of two regions:(i)randomly orientated cellular structure region caused by partially melted powders at the top of each cladding layer;and(ii)epitaxial cellular structure region adjacent to the fusion line.Usually, randomly orientated cellular structure region was totally remelted for a wide range of process conditions. The remelting effect ensures the continuity of epitaxial growth of cellular structure and leads to the formation of columnarβgrains. In order to obtain equiaxed grains the scanning velocity and powder feed rate should be carefully selected to enlarge the randomly orientated cellular structure region. | Qiang Zhang Jing Chen Lilin Wang Hua Tan Xin Lin Weidong Huang | 2016 | Journal of Materials Science & Technology2016,32,4: | 4
The histone methyltransferase EZH2 primes the early differentiation of follicular helper T cells during acute viral infection显示文摘Epigenetic modifications to histones dictate the differentiation of naïve CD4^(+) T cells into different subsets of effector T helper(TH)cells. The histone methyltransferase enhancer of zeste homolog 2(EZH2)has been implicated in the mechanism regulating the differentiation of TH1,TH2 and regulatory T(Treg)cells. However, whether and how EZH2 regulates follicular helper T(TFH)cell differentiation remain unknown.Using a mouse model of acute lymphocytic choriomeningitis virus(LCMV)infection,we observed abundant EZH2 expression and associated H3K27me3 modifications preferentially in the early committed virus-specific TFH cells compared to those in TH1 cells.Ablation of EZH2 in LCMV-specific CD4^(+) T cells leads to a selective impairment of early TFH cell fate commitment, but not late TFH differentiation or memory TFH maintenance.Mechanistically,EZH2 specifically stabilizes the chromatin accessibility of a cluster of genes that are important for TFH fate commitment, particularly B cell lymphoma 6(Bcl6),and thus directs TFH cell commitment. Therefore,we identified the chromatin-modifying enzyme EZH2 as a novel regulator of early TFH differentiation during acute viral infection. | Xiangyu Chen Guoshuai Cao Jialin Wu Xinxin Wang Zhiwei Pan Jianbao Gao Qin Tian Lifan Xu Zhirong Li Yaxing Hao Qizhao Huang Pengcheng Wang Minglu Xiao Luoyingzi Xie Shupei Tang Zhenyu Liu Li Hu Jianfang Tang Ran He Li Wang Xinyuan Zhou Yuzhang Wu Mengjie Chen Beicheng Sun Bo Zhu Jun Huang Lilin Ye | 2020 | Cellular & Molecular Immunology2020,17,3: | 4
The dichotomous and incomplete adaptive immunity in COVID-19 patients with different disease severity显示文摘The adaptive immunity that protects patients from coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is not well characterized. In particular, the asymptomatic patients have been found to induce weak and transient SARS-CoV-2 antibody responses, but the underlying mechanisms remain unknown;meanwhile, the protective immunity that guide the recovery of these asymptomatic patients is elusive. Here,we characterized SARS-CoV-2-specific B-cell and T-cell responses in 10 asymptomatic patients and 64 patients with other disease severity(mild,n=10,moderate,n=32,severe,n=12)and found that asymptomatic or mild symptomatic patients failed to mount virus-specific germinal center(GC)B cell responses that result in robust and prolonged humoral immunity, assessed by GC response indicators including follicular helper T(TFH)cell and memory B cell responses as well as serum CXCL13 levels.Alternatively, these patients mounted potent virus-specific TH1 and CD8+T cell responses. In sharp contrast, patients of moderate or severe disease induced vigorous virus-specific GC B cell responses and associated TFH responses;however, the virus-specific TH1 and CD8+T cells were minimally induced in these patients. These results, therefore, uncovered the protective immunity in asymptomatic patients and also revealed the strikingly dichotomous and incomplete humoral and cellular immune responses in COVID-19 patients with different disease severity, providing important insights into rational design of effective COVID-19 vaccines. | Leiqiong Gao Jing Zhou Sen Yang Lisha Wang Xiangyu Chen Yang Yang Ren Li Zhiwei Pan Jing Zhao Zhirong Li Qizhao Huang Jianfang Tang Li Hu Pinghuang Liu Guozhong Zhang Yaokai Chen Lilin Ye | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 2
Retrorsine Cooperates with Gut Microbiota to Promote Hepatic Sinusoidal Obstruction Syndrome by Disrupting the Gut Barrier显示文摘Background and Aims:Hepatic sinusoidal obstruction syndrome(HSOS)is a life-threatening syndrome, and a cause is exposure to pyrrolizidine alkaloid(PA)-containing products. It is well-established that retrorsine(RTS),a rep-resentative Pas,insults hepatic sinusoidal endothelial cells and ensues congestion of hepatic sinusoids. However, little known about the impact of Pas on gut microbiota and intesti-nal barrier and inflammation in HSOS.Methods:Mice were gavaged with or without nonabsorbable antibiotics(ABX),followed by a single dose of RTS.The gut microbiota was examined by 16S rDNA sequencing. Results:ABX pretreat-ment significantly reversed RTS-induced liver damage.RTS altered gut microbiota composition, increasing Gram-nega-tive bacteria and resulting in a sharp elevation of circulating lipopolysaccharides(LPS)in HSOS mice.Gut decontamina-tion with ABX alleviated RTS-induced intestine inflamma-tion,protected against disruption of the intestinal epithelial barrier and gut vascular barrier(GVB),and suppressed he-patic LPS-NF-κB pathway activation in RTS-induced HSOS.Importantly, the LPS level was positively correlated with MELD score in patients with HSOS.Elevated LPS in patients with HSOS confirmed that Gram-negative bacteria were in-volved in the pathogenesis of HSOS.Conclusions:RTS,a PA,cooperated with gut dysbiosis to cause intestinal inflam-mation and gut barrier compromise that increased transport of gut-derived LPS into the liver through the portal vein, which contributed to the pathology of HSOS.Modulating the gut microbiota, protecting the intestinal barrier, and sup-pressing intestinal inflammation with prebiotics or antibiot-ics might be a useful pharmacologic intervention in HSOS. | Li Xiao Lilin Hu Huikuan Chu Liuying Chen Jingjing Yan Weijun Wang Xiaoqian Yang Qingjing Zhu Fan Du Yuhu Song Peng Chen Xiaohua Hou Ling Yang | 2022 | Journal of Clinical and Translational Hepatology2022,10,6: | 2
Disease severity dictates SARS-CoV-2-specific neutralizing antibody responses in COVID-19显示文摘COVID-19 patients exhibit differential disease severity after SARS-CoV-2 infection. It is currently unknown as to the correlation between the magnitude of neutralizing antibody(NAb)responses and the disease severity in COVID-19 patients. In a cohort of 59 recovered patients with disease severity including severe, moderate, mild, and asymptomatic,we observed the positive correlation between serum neutralizing capacity and disease severity,in particular, the highest NAb capacity in sera from the patients with severe disease, while a lack of ability of asymptomatic patients to mount competent NAbs.Furthermore, the compositions of NAb subtypes were also different between recovered patients with severe symptoms and with mild-tomoderate symptoms. These results reveal the tremendous heterogeneity of SARS-CoV-2-specific NAb responses and their correlations to disease severity, highlighting the needs of future vaccination in COVID-19 patients recovered from asymptomatic or mild illness. | Xiangyu Chen Zhiwei Pan Shuai Yue Fei Yu Junsong Zhang Yang Yang Ren Li Bingfeng Liu Xiaofan Yang Leiqiong Gao Zhirong Li Yao Lin Qizhao Huang Lifan Xu Jianfang Tang Li Hu Jing Zhao Pinghuang Liu Guozhong Zhang Yaokai Chen Kai Deng Lilin Ye | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 1

The gut microbiome and metabolome of Himalayan Griffons(Gyps himalayensis):insights into the adaptation to carrion-feeding habits in avian scavengers显示文摘Background:Himalayan Griffons(Gyps himalayensis),large scavenging raptors widely distributed in Qinghai-Tibetan Plateau, have evolved a remarkable ability to feed on carcasses without suffering any adverse effects. The gut microbiome plays an important role in animal physiological and pathological processes, and has also been found to play a health protective role in the vulture adaptation to scavenging. However, the microbial taxonomic diversity(including nonculturable and culturable microbes),functions, and metabolites related to Himalayan Griffons have not been fully explored. Methods:In the present study, the 28 fecal samples of the Himalayan Griffons and 8 carrion samples were collected and sequenced using high-throughput 16 S rRNA gene sequencing methods to analyze the composition and functional structures of the microbiomes.Twelve fecal samples of the Himalayan Griffons were analyzed using untargeted Liquid Chromatography Mass Spectroscopy(LC-MS)to identify metabolites. We used different culture conditions to grow Himalayan Griffons gut microbes.Inhibitory effects of gut beneficial bacteria on 5 common pathogenic bacteria were also tested using the Oxford cup method. Results:According to the results of the culture-independent method,a high abundance of four major phyla in Himalayan Griffons were identified, including Fusobacteria,Firmicutes,Bacteroidetes, and Proteobacteria. The most abundant genera were Fusobacterium, followed by Clostridium_sensu_stricto_1,Cetobacterium,Epulopiscium,and Bacteroides. The predicted primary functional categories of the Himalayan Griffons'gut microbiome were associated with carbohydrate and amino acid metabolism, replication and repair, and membrane transport.LC-MS metabolomic analysis showed a total of 154 metabolites in all the fecal samples.Cultivation yielded 184 bacterial isolates with Escherichia coli,Enterococcus faecium,Enterococcus hirae, and Paeniclostridium sordellii as most common isolates.Moreover,7 potential beneficial gut bacteria isolated showed certain inhibition to 5 common pathogenic bacteria. Conclusions:Our findings broaden and deepen the understanding of Himalayan Griffons'gut microbiome, and highlighted the importance of gut microbiome-mediated adaptation to scavenging habits. In particular, our results highlighted the protective role of gut beneficial bacteria in the Himalayan Griffons against pathogenic bacteria that appear in rotten food resources. | Wen Wang Xiaolong Gao Sisi Zheng Zhuoma Lancuo Ying Li Lilin Zhu Jianping Hou Jiayi Hai Xin Long Hanxi Chen Alexey Druzyaka Kirill Sharshov | 2021 | Avian Research2021,12,4: | 1
A CSCW SYSTEM FOR BUILDING REVIEWING BY INTEGRATING GIS WITH OA显示文摘The process of urban building reviewing is a collaborative work by a group of urban planning staffs and land managers who are working with different departments of a or- ganization. There are three ingredients for improving the work efficiency, i. e.,agile commu- nication and interactive mechanisms, good information, effective processing and visualizing tools. In order to build a computer supported collaborative work (CSCW) system in Urban Planning and Land Administration Bureau in Changzhou Municipality, GIS was integrated with office automation (OA). The workflow and organizational structure had been re-engi- neered to meet the requirement of application of the new technology. Federated databases were constructed by assembling disparate sources of heterogeneous spatial and attribute data. Easy access to the data by contents was developed for promoting information sharing within the organization. Specific desktop spatial data handling tools were provided for delineating 1and lots or building layouts on large-scale digital maps. Office automation functions, includ- ing applications registering, reasonableness checking, opinions giving and exchanging, were developed and they permit printing and documents filing. The work efficiency in the organi- zation has been improved by introducing the computer-based collaborative building reviewing system. | JIANG Jie CHEN Jun YAN Ronghua XU Lilin | 2000 | Geo-Spatial Information Science2000,3,1: | 1
Efficient control of chronic LCMV infection by a CD4 T cell epitope-based heterologous prime-boost vaccination in a murine model显示文摘CD4^(+)T cells are essential for sustaining CD8^(+)T cell responses during a chronic infection. The adoptive transfer of virus-specific CD4^(+)T cells has been shown to efficiently rescue exhausted CD8^(+)T cells. However, the question of whether endogenous virus-specific CD4^(+)T cell responses can be enhanced by certain vaccination strategies and subsequently reinvigorate exhausted CD8^(+)T cells remains unexplored. In this study,we developed a CD4^(+)T cell epitope-based heterologous prime-boost immunization strategy and examined the efficacy of this strategy using a mouse model of chronic lymphocytic choriomeningitis virus(LCMV)infection. We primed chronically LCMV-infected mice with a Listeria monocytogenes vector that expressed the LCMV glycoprotein-specific I-Ab-restricted CD4^(+)T cell epitope GP61–80(LM-GP61)and subsequently boosted the primed mice with an influenza virus A(PR8 strain)vector that expressed the same CD4^(+)T cell epitope(IAV-GP61).This heterologous prime-boost vaccination strategy elicited strong anti-viral CD4^(+)T cell responses, which further improved both the quantity and quality of the virusspecific CD8^(+)T cells and led to better control of the viral loads. The combination of this strategy and the blockade of the programmed cell death-1(PD-1)inhibitory pathway further enhanced the anti-viral CD8^(+)T cell responses and viral clearance. Thus,a heterologous prime-boost immunization that selectively induces virus-specific CD4^(+)T cell responses in conjunction with blockade of the inhibitory pathway may represent a promising therapeutic approach to treating patients with chronic viral infections. | Ran He Xinxin Yang Cheng Liu Xiangyu Chen Lin Wang Minglu Xiao Jianqiang Ye Yuzhang Wu Lilin Ye | 2018 | Cellular & Molecular Immunology2018,15,9: | 1

A new optimization algorithm applied in electromagnetics——Maxwell’s equations derived optimization(MEDO)显示文摘In this paper,a novel global optimization algorithm, named as Maxwell’s equations derived optimization(MEDO),is proposed.Using the Maxwell’s equations to analyse the behaviors of the timevarying current, the Ampere force is obtained from Fleming’s left hand rule.MEDO introduces an‘Ampere force’term, which is derived from Maxwell’s equations and is rigorous in physics,to drive the variables to the global optimal solution in the search space. In addition, introducing‘gravity’to MEDO can increase the stability of the optimizations.11 classical benchmarks are tested, and results show that MEDO can always converge to numerical optimal solutions. To evaluate the proposed MEDO in solving the electromagnetic problems, four practical engineering applications are considered including the linear antenna array synthesis, frequency selected surface optimization, numerical dispersion reduction for finite-difference method, and parameters extraction of typical waveform. These examples are significant in electromagnetics, but tough to be solved because of their high dimensionality and strong nonlinearity.Numerical results show that MEDO can outperform several classic optimization methods, like wind driven optimization(WDO)and particle swarm optimization(PSO).Therefore, the electromagnetics-inspired MEDO is robust and of great potential in solving the electromagnetic optimization problems. | Donglin SU Lilin LI Shunchuan YANG Bing LI Guangzhi CHEN Hui XU | 2020 | Science China(Information Sciences)2020,63,10: | 1
