2New incompatible pair of TCM:Epimedii Folium combined with Psoraleae Fructus induces idiosyncratic hepatotoxicity under immunological stress conditions显示文摘Epimedii Folium(EF)combined with Psoraleae Fructus(PF)is a common modern preparation,but liver injury caused by Chinese patent medicine preparations containing EF and PF has been frequently reported in recent years.Zhuangguguanjiewan pills(ZGW),which contain EF and PF,could induce immune idiosyncratic liver injury according to clinical case reports and a nonhepatotoxic dose of lipopolysaccharide(LPS)model.This present study evaluated the liver injury induced by EF or PF alone or in combination and investigated the related mechanism by using the LPS model.Liver function indexes and pathological results showed that either EF or PF alone or in combination led to liver injury in normal rats;however,EF or PF alone could lead to liver injury in LPS-treated rats.Moreover,EF combined with PF could induce a greater degree of injury than that caused by EF or PF alone in LPS-treated rats.Furthermore,EF or PF alone or in combination enhanced the LPS-stimulated inflammatory cytokine production,implying that IL-1β,which is processed and released by activating the NLRP3 inflammasome,is a specific indicator of EF-induced immune idiosyncratic hepatotoxicity.Thus,EF may induce liver injury through enhancing the LPS-mediated proinflammatory cytokine production and activating the NLRP3 inflammasome.In addition,the metabolomics analysis results showed that PF affected more metabolites in glycerophospholipid and sphingolipid metabolic pathways compared with EF in LPS model,suggesting that PF increased the responsiveness of the liver to LPS or other inflammatory mediators via modulation of multiple metabolic pathways.Therefore,EF and PF combination indicates traditional Chinese medicine incompatibility,considering that it induces idiosyncratic hepatotoxicity under immunological stress conditions.Yuan Gao Zhilei Wang Jinfa Tang Xiaoyi Liu Wei Shi Nan Qin Xiaoyan Wang Yu Pang Ruisheng Li Yaming Zhang Jiabo Wang Ming Niu Zhaofang Bai Xiaohe Xiao 2020Frontiers of Medicine2020,14,1:32
3Characterizations and mechanical properties of impregnated diamond segment using Cu-Fe-Co metal matrix显示文摘Diamond impregnated Cu-Fe-Co based saw-blade segments are directly processed by vacuum and pressure-assisted sintering at different temperature,with the purpose of reducing the cobalt content in diamond tools.Copper and iron are used as the bonding elements and co-balt-chrome pre-alloyed powder is used as the hardening phase.Effects of sintering temperature on microstructures and mechanical properties of the sintered matrix and diamond graphitization were investigated by X-ray diffraction analysis,electron probe micro-analyzer,universal testing machine,digital Rockwell hardness tester and Raman scattering analyzer.Results showed that microstructures of the sintered matrix were refined and porosities in the sintered matrix were closed to a more spherical-like shape with the increase of the sintering temperature.Densification,hardness and tensile strength of the matrix sintered at 820 ℃ were 12.75%,2.72% and 156.38% higher than that of the matrix sintered at 740 ℃,respectively.Diamond graphitization was not occurred at 820 ℃.The hardness and the tensile strength rose 32.8% and 13.5%,respectively,after 7.5 h ageing treatment.The matrix densification ascent and the dispersed distribution of Co-Cr pre-alloyed powders contributed a hardness improvement and a tensile strength improvement to the Cu-Fe based matrix.Li, Wensheng Zhang, Jie Wang, Shucai Dong, Hongfeng Li, Yaming Liu, Yi 2012Rare Metals2012,31,1:19
4Uplift and evolution of Helan Mountain显示文摘The Helan Mountain lies in the northwest margin of Ordos Basin and its uplift periods have close relations with the tectonic feature and evolution of the basin. There are many views on the uplift time of Helan Mountain, which is Late Triassic and Late Jurassic. It is concluded by the present strata, magmatic rock and hot fluid distribution that the Helan Mountain does not uplift in Late Triassic to Middle Jurassic but after Middle Jurassic. Through the research of the sedimentary strata and deposit rate in Yinchuan Graben which is near to the Helan Mountain, it is proved that the Helan Mountain uplifts in Eocene with a huge scale and in Pliocene with a rapid speed. The fission track analysis of apatite and zircon can be used to determine the precise uplift time of Helan Mountain, which shows that four stages of uplifting or cooling: Late Jurassic to the early stage of Early Cretaceous, mid-late stage of Early Cretaceous, Late Cretaceous and since Eocene. During the later two stages the uplift is most apparent and the mid-late stage of Early Cretaceous is a regional cooling course. 5A Simulator for Producing of High Flux Atomic Oxygen Beam by Using ECR Plasma Source显示文摘In order to study the atomic oxygen corrosion of spacecraft materials in low earth orbit environment, an atomic oxygen simulator was established. In the simulator, a 2.45 GHz microwave source with maximum power of 600 W was launched into the circular cavity to generate ECR (electron cyclotron resonance) plasma. The oxygen ion beam moved onto a negatively biased Mo plate under the condition of symmetry magnetic mirror field confine, then was neutralized and reflected to form oxygen atom beam. The properties of plasma density, electron temperature, plasma space potential and ion incident energy were characterized. The atomic oxygen beam flux was calibrated by measuring the mass loss rate of Kapton during the atomic 5~30 eV and a cross section of φ80 mm could be obtained under the operating pressure of 10-1~10-3 Pa. 7The Role of Host Cytoskeleton in Flavivirus Infection显示文摘The family of flaviviruses is one of the most medically important groups of emerging arthropod-borne viruses. Host cell cytoskeletons have been reported to have close contact with flaviviruses during virus entry, intracellular transport, replication, and egress process, although many detailed mechanisms are still unclear. This article provides a brief overview of the function of the most prominent flaviviruses-induced or-hijacked cytoskeletal structures including actin, microtubules and intermediate filaments, mainly focus on infection by dengue virus, Zika virus and West Nile virus. 8New insights into hepatitis B virus biology and implications for novel antiviral strategies显示文摘Hepatitis B virus(HBV), a small DNA virus with a unique replication mode, can cause chronic hepatitis(CHB), which is characterized by the persistence of the viral covalently closed circular DNA that serves as the template for HBV replication and the production of large amounts of secreted HBV surface antigen(HBs Ag) that is present in excess of the levels of infectious virus. Despite the success of currently approved antiviral treatments for CHB patients, including interferon and nucleotide analogs, which suppress HBV replication and reduce the risk of CHB-related liver diseases, these therapies fail to eradicate the virus in most of the patients. With the development of the cell and animal models for HBV study, a beter understanding of the HBV life cycle has been achieved and a series of novel antiviral strategies that target diferent stages of HBV replication have been designed to overcome the viral factors that contribute to HBV persistence. 9Sandwich ELISA for detecting urinary Survivin in bladder cancer显示文摘Objective: Survivin as a tumor marker in the diagnosis of bladder cancer has not been completely confirmed yet and there are few reports about using Survivin enzyme-linked immunosorbent assay (ELISA) kit to detect the urine of bladder cancer patients. This study aimed to develop a Survivin ELISA and validate its value in the detection of bladder cancer. Methods: Through square matrix titration, different combinations of coating antibody and detecting antibody, a Survivin ELISA was constructed. This assay was evaluated according to intra-assay precision, inter-assay precision and minimum detectable dose (MDD). Survivin levels were detected and analyzed in 102 bladder cancer patients and 102 healthy people by established ELISA. Then cutoff value was defined according to the analysis of receiver operating characteristic (ROC) curve. The sensitivity and specificity of detection were calculated on the basis of cutoff value to diagnose bladder cancer patients. Furthermore, the value of Survivin expression detected by ELISA among different clinicopathological characteristics of patients was also compared. Results: Through optimization of different conditions, intra-assay precision was 8.39%, inter-assay precision 8.57% and MDD 0.0625 ng/mL in this assay. When the optical density at 450 nm (OD 450 ) was 0.09, it could get the optimized diagnostic cutoff value. According to this value, the sensitivity and specificity of diagnosis in bladder cancer patients were 70.6% and 89.2%, respectively. The associations between patients' clinical variables and OD 450 were not significant except tumor numbers in patients. 11Hybrid Structure Reliability Analysis Based on the Damped Newton Method显示文摘This paper presents a hybrid model reliability analysis method based on the damped Newton method with both random and interval variables to solve the hybrid structure reliability problem.The method combines an outer iterative solution and inner layer numerical calculation.In the outer iteration,the method seeks an optimized solution to the interval variable iterative by adding the boundary constraint condition based on the damped Newton optimization theory.In the inner layer solution,the method first reduces the dimension of the random variable through the dimension reduction method,then obtains the first four-order central moment of the function through the application of the Taylor expansion method,and finally calculates the reliability index of the structure according to the fourth-order moment calculation structure of the function.The results of a numerical example and an engineering ten-rod truss structure show that the proposed method can effectively solve the random-interval hybrid reliability problem and has better calculation accuracy than that of the two-layer iterative method.Hongwei Zheng Guangwei Meng Feng Li Tonghui Wei Wei Luo Yaming Guo 2020Tsinghua Science and Technology2020,25,5:2

13Dynamics and manipulation of ferroelectric domain walls in bismuth ferrite thin films显示文摘Ferroelectric domain walls differ from domains not only in their crystalline and discrete symmetry,but also in their electronic,magnetic,and mechanical properties.Although domain walls provide a degree of freedom to regulate the physical properties at the nanoscale;the relatively lower controllability prevents their practical applications in nano-devices.In this work;with the advantages of 3D domain configuration detection based on piezoresponse force microscopy;we find that the mobility of three types of domain walls(tail-to-tail,head-to-tail,head-to-head)in(001)BiFeO3 films varies with the applied electrical field.Under low voltages;head-to-tail domain walls are more mobile than other domain walls;while,under high voltages,tail-to-tail domain walls become rather active and possess relatively long average lengths.This is due to the high nucleation energy and relatively low growth energy for charged domain walls.Finally;we demonstrate the manipulation of domain walls through successive electric writings,resulting in well-aligned conduction paths as designed;paving the way for their application in advanced spintronic;memory and communication nano-devices.Shuyu Xiao Yaming Jin Xiaomei Lu Sang-Wook Cheong Jiangyu Li Yang Li Fengzhen Huang Jinsong Zhu 2020National Science Review2020,7,2:2

15Lineage tracing reveals the origin of Nestin-positive cells are heterogeneous and rarely from ependymal cells after spinal cord injury显示文摘Nestin is expressed extensively in neural stem/progenitor cells during neural development, but its expression is mainly restricted to the ependymal cells in the adult spinal cord. After spinal cord injury(SCI), Nestin expression is reactivated and Nestinpositive(Nestin;) cells aggregate at the injury site. However, the derivation of Nestin;cells is not clearly defined. Here, we found that Nestin expression was substantially increased in the lesion edge and lesion core after SCI. Using a tamoxifen inducible CreER(T2)-loxP system, we verified that ependymal cells contribute few Nestin;cells either to the lesion core or the lesion edge after SCI. In the lesion edge, GFAP+astrocytes were the main cell type that expressed Nestin;they then formed an astrocyte scar.In the lesion core, Nestin;cells expressed αSMA or Desmin, indicating that they might be derived from pericytes. 16Major Tectono-thermal Events in the Yangtze Craton: Insights from U-Pb-Lu-Hf Isotope Records in Zircons from End-Permian Volcanic Interlayers in Southwest China显示文摘The in situ zircon U-Pb-Lu-Hf isotope records from end-Permian volcanic interlayers in southwest China,integrated with previous studies,restructure the evolutionary history of the Yangtze Craton from Precambrian to Late Paleozoic.This includes early continental crust formation before 3.0 Ga and massive juvenile crustal growth at 2.6-2.4 Ga;large-scale crustal reworking at 2.1-1.7 Ga;Neoproterozoic crust addition at 1.1 to 0.7 Ga;collision and subduction along the craton margin between 700-541 Ma;Early Ordovician to Late Silurian magmatism;and large tectono-thermal events in the Middle Carboniferous to end-Permian.Some zircons with T(MD2)ages from 4.40 to 4.01 Ga and lower initial176Hf/177Hf values of 0.280592 to 0.280726 may imply the existence of Hadean crust relics beneath the Yangtze Craton and their provenances could be associated with Hadean crustal remelting.This study further clarifies that the Precambrian-age zircons between the end-Permian volcanic interlayers,the complexes in the western margin of the Yangtze Craton,and the sedimentary Kangdian Basin,may share an affinity based on similar U-Pb age spectra and Hf isotope features.It also shows that the Neoproterozoic tectono-thermal event may be associated with large-scale tectono-rifting activity,which is different from the Grenville-age continental collision between Yangtze and Cathaysia blocks in South China.The above findings support the inference of a widespread Archean basement extending to the western Yangtze Craton and a provenance in the Kangdian Basin that is derived from the weathering and erosion of Paleoproterozoic continental crust.SHI Zejin LI Wenjie ZHANG Jin TIAN Yaming WANG Yong YIN Guan LUO Qichao 2020Acta Geologica Sinica(English Edition)2020,94,6:1
17CircRNA.0007127 triggers apoptosis through the miR-513a-5p/CASP8 axis in K-562 cells显示文摘Background:Circular RNAs(circR NAs)are covalently closed single-stranded RNAs with multiple biological functions.CircRNA.0007127 is derived from the carbon catabolite repression 4-negative on TATA-less(CCR4-NOT)complex subunit2(CNOT2),which was found to regulate tumor cell apoptosis through caspase pathway.Methods:Potential circR NA.0007127 target microRNAs(miRNAs)were analyzed by miRanda,TargetScan,and RNAhybrid software,and the miRNAs with binding sites for apoptosis-related genes were screened.The roles of circR NA.0007127 and its downstream target,microR NA(miR)-513a-5p,were validated by quantitative real-time polymerase chain reaction(qPCR),flow cytometry,mitochondrial membrane potential,immunofluorescence,western blot,and caspase-8(CASP8)protein activity in vitro in HO-induced K-562 cells.The circRNA.0007127-miR-513a-5p and CASP8-miR-513a-5p interactions were verified by luciferase reporter assays.Results:Silencing circRNA.0007127 decreased cell apoptosis by inhibiting CASP8 pathway activation in K-562 cells.Compared with the control group,the expression of CASP8 was reduced by 50%and the 43-kD fragment of CASP8 protein was significantly reduced(P≤0.05).The luciferase reporting assay showed that circRNA.0007127 combined with miR-513a-5p or CASP8,with extremely significant differences(P≤0.001).The overexpression of miR-513a-5p inhibited the gene expression level of CASP8in a human myeloid leukemia cell model(75%change)and the level of a 43-kD fragment of CASP8 protein(P small-interfering RNA(siRNA)and the miR-≤0.01).The rescue experiment showed that cotransfection with circRNA.0007127513a-5p inhibitor increased CASP8 gene expression and the apoptosis rate,suggesting that the miR-513a-5p inhibitor is a circRNA.0007127siRNA antagonist.Conclusions:CircRNA.0007127 regulates K-562 cell apoptosis through the miR-513a-5p/CASP8 axis,which can serve as a novel powerful molecular target for K-562 cells.Xiajing LI Yiyu ZHANG Ning WANG Zhaohu YUAN Xiaojie CHEN Qicong CHEN Hui DENG Xinxin TONG Honglin CHEN Yuyou DUAN Yaming WEI 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,9:1
18Ultrafine Fe-modulated Ni nanoparticles embedded within nitrogen-doped carbon from Zr-MOFs-confined conversion for efficient oxygen evolution reaction显示文摘Improvement of the low-cost transition metal electrocatalyst used in sluggish oxygen evolution reaction is a significant but challenging problem. In this study, ultrafine Fe-modulated Ni nanoparticles embedded in a porous Ni-doped carbon matrix were produced by the pyrolysis of zirconium metal–organic–frameworks, in which 2,2′-bipyridine-5,5′-dicarboxylate operating as a ligand can coordinate with Ni^(2+) and Fe^(3+). This strategy allows formation of Fe-modulated Ni nanoparticles with a uniform dimension of about 2 nm which can be ascribed to the spatial blocking effect of ZrO_(2). This unique catalyst displays an efficient oxygen evolution reaction electrocatalytic activity with a low overpotential of 372 mV at 10 mA·cm^(–2) and a small Tafel slope of 84.4 mV·dec^(–1) in alkaline media. More importantly, it shows superior durability and structural stability after 43 h in a chronoamperometry test. Meanwhile, it shows excellent cycling stability during 4000 cyclic voltammetry cycles.
