2Construction of Prokaryotic Expression Vector of Mouse Nanog Gene and Its Expression显示文摘The aim of this study is to construct a prokaryotic expression vector of mouse Nanog gene and to express it in E.coli.A pair of primers was designed according to digestion sites in plasmid pGEX-KG and the Nanog gene sequence published by GenBank.The DNA fragment of 918 bp was amplified by polymerase chain reaction(PCR)from the pNA992 recombinant plasmid with Nanog gene,then cloned into pGEX-KG and transformed into the host E.coli strain TGⅠ.The sequence of the fragment was matched with the original sequence of pNA992.It indicated that fusion expression vector,pGEX-KG- Nanog,was constructed successfully.The pGEX-KG-Nanog plasmid was extracted from E.coli strain TGⅠand was transformed into BL21(DE3)for expression.After induction by isopropy1-β-D-thiogalactoside(IPTG)at 37°C,the expression product of Nanog gene was identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),and the expression condition was optimized.Nanog fusion protein was successfully expressed in the form of inclusion bodies. The molecular weight of the inclusion body was 63 kDa.Meanwhile,the optimum condition for the expression of Nanog fusion protein was induced with 0.8 mmol L-1 IPTG for 5 h.The mouse Nanog gene was successfully expressed in E.coli, which laid a foundation for the purification of Nanog protein and for the preparation of polyclonal antibody.LI Jun Lü Chang-rong DOU Lin DOU Zhong-ying 2007Agricultural Sciences in China2007,6,4:3

5Probabilistic assessment of tunnel convergence considering spatial variability in rock mass properties using interpolated autocorrelation and response surface method显示文摘This study aims at the probabilistic assessment of tunnel convergence considering the spatial variability in rock mass properties. The method of interpolated autocorrelation combined with finite difference analysis is adopted to model the spatial variability of rock mass properties. An iterative procedure using the first-order reliability method(FORM) and response surface method(RSM) is employed to compute the reliability index and its corresponding design point. The results indicate that the spatial variability considerably affects the computed reliability index. The probability of failure could be noticeably overestimated in the case where the spatial variability is neglected. The vertical scale of fluctuation has a much higher effect on the probabilistic result with respect to the tunnel convergence than the horizontal scale of fluctuation. 6Recursive formulations for the method of reverberation-ray matrix and the application显示文摘A recursive formulation is proposed for the method of reverberation-ray matrix (MRRM) to exactly analyze the free vibration of a multi-span continuous rectangular Kirchhoff plate, which has two oppo- site simply-supported edges. In contrast to the traditional MRRM, numerical stability is achieved by using the present new formulation for high-order frequencies or/and for plates with large span-to-width ratios. The heavy computational cost of storage and memory are also cut down. An improved recursive formulation is further proposed by modifying the iterative formula to reduce the matrix inversion op- erations. 7Predicting the Nuclear Localization Signals of 107 Types of HPV L1 Proteins by Bioinformatic Analysis显示文摘In this study, 107 types of human papillomavirus (HPV) L1 protein sequences were obtained from available databases, and the nuclear localization signals (NLSs) of these HPV L1 proteins were analyzed and predicted by bioinformatic analysis. Out of the 107 types, the NLSs of 39 types were predicted by PredictNLS soft- ware (35 types of bipartite NLSs and 4 types of monopartite NLSs). The NLSs of the remaining HPV types were predicted according to the characteristics and the homology of the already predicted NLSs as well as the general rule of NLSs. According to the result, the NLSs of 107 types of HPV L1 proteins were classified into 15 categories. The different types of HPV L1 proteins in the same NLS cat- egory could share the similar or the same nucleocytoplasmic transport pathway. They might be used as the same target to prevent and treat different types of HPV infection. 8Mobile node aware opportunistic routing in dynamic wireless sensor network显示文摘Opportunistic routing(OR) could adapt to dynamic wireless sensor networks(WSNs) because of its inherent broadcast nature.Most of the existing OR protocols focus on the variations of propagation environment which are caused by channel fading.However, a few works deal with the dynamic scenario with mobile nodes.In this paper, a mobile node(MN) aware OR(MN-OR) is proposed and applied to a WSN in the high-speed railway scenario where the destination node is deployed inside a high speed moving train, and the MN-OR not only considers the mobility of node but also utilizes the candidate waiting time induced by the timer-based coordination scheme.Specifically, to reduce the number of duplicate transmissions and mitigate the delay of information transmission, a new selection strategy of the candidate forwarders is presented.In addition, two priority assignment methods of the candidate forwarders are proposed for the general relay nodes(GRNs) and the rail-side nodes(RSNs) according to their different routing requirements.Extensive simulation results demonstrate that the proposed MN-OR protocol can achieve better network performances compared with some existing routing schemes such as the well-known Ad-hoc on-demand distance vector routing(AODV) and the extremely opportunistic routing(ExOR) protocols.Lü Xiaojun Jia Xinchun Han Zongyuan Yang Bo Hao Jun 2016The Journal of China Universities of Posts and Telecommunications2016,23,5:1

12Fast ISAR imaging method based on scene segmentation显示文摘Although compressed sensing inverse synthetic aperture radar(ISAR) imaging methods are widely used in radar signal processing, its reconstructing time and memory storage space requirements are very high. The main reason is that large scene reconstruction needs a higher dimension of the sensing matrix. To reduce this limitation, a fast high resolution ISAR imaging method,which is based on scene segmentation for random chirp frequencystepped signals, is proposed. The idea of scene segmentation is used to solve the problems aforementioned. In the method,firstly, the observed scene is divided into multiple sub-scenes and then the sub-scenes are reconstructed respectively. Secondly, the whole image scene can be obtained through the stitching of the sub-scenes. Due to the reduction of the dimension of the sensing matrix, the requirement of the memory storage space is reduced substantially. In addition, due to the nonlinear superposition of the reconstructed time of the segmented sub-scenes, the reconstruction time is reduced, and the purpose of fast imaging is achieved.Meanwhile, the feasibility and the related factors which affect the performance of the proposed method are also analyzed, and the selection criterion of the scene segmentation is afforded. 13Bioinformatics-led design of single-chain antibody molecules targeting DNA sequences for retinoblastoma显示文摘The development of single-chain Fv antibody(scFv) by recombinant gene expression is an important milestone for cancer therapy.Single-chain antibodies are reconstructed for cancer-targeted therapy to provide good penetration into tumor tissue and to improve their pharmacokinetics in vivo,offering a clinically valuable application.The relationship needs to be analyzed that there may be some variations between the structure and function of the fusion proteins,and the relationship between the structure and function of protein molecules was obtained through analyzing relevant literature at home and abroad as well as modeling analysis.Through our analysis of the interaction region between antibody and antigen,and of the binding sites for molecular conformation,it is clear that existing antibodies need to be modified at the DNA sequence level,enhancing the biological activity of the antibodies.Based on the view that bio-molecular computer models are closely integrated with biological experiments,a bio-molecular structure-activity relationship model can be established in terms of molecular conformation,physical and chemical properties and the biological activity of single-chain antibodies.Two enlightenments are obtained from our analysis.On one hand,the structure-activity relationship is clear for new immune molecules at the gene expression level.On the other hand,a single-chain antibody molecule can be designed and optimized for the cancer-oriented treatment.In this article,we provided the theoretical and experimental basis for the development of single-chain antibodies appropriate for retinoblastoma therapy.Guo-Gang Shang, Jun Yun 2011International Journal of Ophthalmology(English edition)2011,4,1:1

15Synchrotron infrared spectral regions as signatures for foodborne bacterial typing显示文摘Fourier-transform infrared(FTIR) spectroscopy has emerged as a viable alternative to biochemical and molecular biology techniques for bacterial typing with advantages such as short analysis time, low cost and laboratorial simplicity. In this study, synchrotron radiationbased FTIR(SR-FTIR) spectroscopy with higher spectral quality was successfully applied to type 16 foodborne pathogenic bacterial strains. Combined with principal component analysis(PCA) and hierarchical cluster analysis(HCA), we found that the specific spectral region1300-1000 cm^(-1), which reflects the information of phosphate compounds and polysaccharides, can be used as the signature region to cluster the strains into groups similar with genetic taxonomic method. 17Investigation of temperature-dependent small-signal performances of TB SOI MOSFETs显示文摘This paper investigated the temperature dependence of the cryogenic small-signal ac performances of multi-finger partially depleted(PD) silicon-on-insulator(SOI) metal oxide semiconductor field effect transistors(MOSFETs),with T-gate body contact(TB) structure.The measurement results show that the cut-off frequency increases from 78 GHz at 300 K to 120 GHz at 77 K and the maximum oscillation frequency increases from 54 GHz at 300 K to 80 GHz at 77 K,and these are mainly due to the effect of negative temperature dependence of threshold voltage and transconductance.By using a simple equivalent circuit model,the temperature-dependent small-signal parameters are discussed in detail.The understanding of cryogenic small-signal performance is beneficial to develop the PD SOI MOSFETs integrated circuits for ultra-low temperature applications.Yuping Huang Jun Liu Kai Lü Jing Chen 2017Journal of Semiconductors2017,38,4:0

19Telecom Regulation of Next Generation Network in China显示文摘The advent of the Next Generation Network (NGN), a new service-driven network, urges the telecom service operators to consider transforming from single-service providers to full-service providers. During the transformation, they should be concerned about the network user number and the network quality as well as the value added network information. The low threshold for service provision brings a new breed of service providers, which impacts upon the current regulation policy. To adapt to the development of the NGN, it is a necessity to improve the regulation policy in terms of service operators management, user management, Quality of Service (QoS) assurance, service monitoring, charging, and settlement. Meanwhile, regulatory authorities should establish a new body as quickly as possible to meet the trend of the NGN convergence. 20Estimation of fracture size and azimuth in the universal elliptical disc model based on trace information显示文摘The geometric characteristics of fractures within a rock mass can be inferred by the data sampling from boreholes or exposed surfaces.Recently,the universal elliptical disc(UED)model was developed to represent natural fractures,where the fracture is assumed to be an elliptical disc and the fracture orientation,rotation angle,length of the long axis and ratio of short-long axis lengths are considered as variables.This paper aims to estimate the fracture size-and azimuth-related parameters in the UED model based on the trace information from sampling windows.The stereological relationship between the trace length,size-and azimuth-related parameters of the UED model was established,and the formulae of the mean value and standard deviation of trace length were proposed.The proposed formulae were validated via the Monte Carlo simulations with less than 5%of error rate between the calculated and true values.With respect to the estimation of the size-and azimuth-related parameters using the trace length,an optimization method was developed based on the pre-assumed size and azimuth distribution forms.A hypothetical case study was designed to illustrate and verify the parameter estimation method,where three combinations of the sampling windows were used to estimate the parameters,and the results showed that the estimated values could agree well with the true values.Furthermore,a hypothetical three-dimensional(3D)elliptical fracture network was constructed,and the circular disc,non-UED and UED models were used to represent it.The simulated trace information from different models was compared,and the results clearly illustrated the superiority of the proposed UED model over the existing circular disc and non-UED models。Jichao Guo Jun Zheng Qing Lü Jianhui Deng 2023Journal of Rock Mechanics and Geotechnical Engineering2023,15,6:0
