| 1 | CTLA4 Silencing with siRNA Promotes Deviation of Th1/Th2 in Chronic Hepatitis B Patients显示文摘To determine whether RNA interference (RNAi) could block cytotoxic T-lymphocyte antigen 4 (CTLA4) in human lymphocytes in vitro and promote IFN-γ and IL-2 secretions, three small interfering RNAs (siRNAs) were selected based on target specificity sequences of human CTLA4 and transfected into human lymphocytes of chronic HBV patients. As a result, the expression of human CTLA4 mRNA was efficiently suppressed by all the three siRNAs. Compared with negative control (siRNA-co), siRNA-1 inhibited the expression of CTLA4 most efficiently and was used in the further study. The expressions of IFN-γ and IL-2 were upregulated and the level of IL-4 was almost unchanged in lymphocytes transfected with siRNA-1 compared with the blank control. These results indicated that siRNA-1 led to IFN-γ and IL-2 secretions, which is a main response of Th1/Th2. In a conclusion, RNAi significantly suppressed the expression of human CTLA4 mRNA in human lymphocytes in vitro, and could induce Th1/Th2 response. | 4 | Response of the sediment geochemistry of the Changjiang River(Yangtze River) to the impoundment of the Three Gorges Dam显示文摘Based on the measurement of major and trace elements in suspended sediments in the low reaches of the Changjiang River during throughout a whole hydrologic year, the origins, seasonal variations, and fluxes of multielements and the human impacts on multielements transport processes have been analyzed along with the influence of weathering in the Changjiang River basin. The results show that most element contents were high in both autumn and winter and low in summer, which was largely caused by the dilution of discharge. Weathering detritus in the Changjiang River basin is the main source of most elements in suspended sediments. However, riverine pollution could bring more loadings of Cd, Pb, As and Zn into river water than it did a few decades ago. The annual average fluxes of Cd, Pb and Zn, which are major contamination elements, to the sea were 179 ± 21 tons/year, 7810 ± 675 tons/year, and 12,000 ± 1320 tons/year,respectively, in which approximately 8.7%, 11.9% and 2.7% of their loadings, respectively,were contributed by pollution inputs. Element exports mainly occurred in the summer(44.4%–57.4%) in the lower part of the Changjiang River. A general relationship between sediment retention and element content suggests a positive feedback mechanism for the decreased number of particles, in which element riverine loadings are reduced due to the enhanced trapping effect by the Three Gorges Dam(TGD). | 5 | Dielectric metalens for miniaturized imaging systems:progress and challenges显示文摘Lightweight, miniaturized optical imaging systems are vastly anticipated in these fields of aerospace exploration, industrial vision, consumer electronics, and medical imaging. However, conventional optical techniques are intricate to downscale as refractive lenses mostly rely on phase accumulation. Metalens, composed of subwavelength nanostructures that locally control light waves, offers a disruptive path for small-scale imaging systems. Recent advances in the design and nanofabrication of dielectric metalenses have led to some high-performance practical optical systems. This review outlines the exciting developments in the aforementioned area whilst highlighting the challenges of using dielectric metalenses to replace conventional optics in miniature optical systems. After a brief introduction to the fundamental physics of dielectric metalenses, the progress and challenges in terms of the typical performances are introduced. The supplementary discussion on the common challenges hindering further development is also presented, including the limitations of the conventional design methods, difficulties in scaling up, and device integration. | 7 | Transgenic Arabidopsis thaliana expressing a wheat oxalate oxidase exhibits hydrogen peroxide related defense response显示文摘Oxalic acid(OA) is considered as an important pathogenetic factor of some destructive diseases caused by some fungal pathogens such as Sclerotinia sclerotiorum. Oxalate degradation is important for plant health, and plants that contain oxalate oxidase(OXO) enzymes could breakdown oxalate into CO_2 and H_2O_2, which subsequently evokes defense responses. However, some species, such as Arabidopsis thaliana, have no oxalate oxidase activity identified to date. The present study aims to develop transgenic Arabidopsis expressing a wheat oxalate oxidase, to test for the response to OA exposure and fungal infection by S. sclerotiorum. The results showed that the transgenic Arabidopsis lines that expressed the wheat OXO exhibited enhanced resistance to OA exposure and S. sclerotiorum infection in the tolerance assays. In the same manner, it could convert OA to CO_2 and H_2O_2 to a higher extent than the wild-type. Intensive osmotic adjustments were also detected in the transgenic Arabidopsis lines. The higher level of produced H_2O_2 subsequently induced an elevated activity of antioxidant enzymes including superoxide dismutase(SOD) and peroxidase(POD) in the transgenic Arabidopsis plants. | 10 | Phosphorylation of PHF2 by AMPK releases the repressive H3K9me2 and inhibits cancer metastasis显示文摘Epithelial to mesenchymal transition (EMT) plays a crucial role in cancer metastasis, accompanied with vast epigenetic changes.AMP-activated protein kinase (AMPK), a cellular energy sensor, plays regulatory roles in multiple biological processes. Although afew studies have shed light on AMPK regulating cancer metastasis, the inside epigenetic mechanisms remain unknown. Herein weshow that AMPK activation by metformin relieves the repressive H3K9me2-mediated silencing of epithelial genes (e.g., CDH1)during EMT processes and inhibits lung cancer metastasis. PHF2, a H3K9me2 demethylase, was identified to interact with AMPKα2.Genetic deletion of PHF2 aggravates lung cancer metastasis and abolishes the H3K9me2 downregulation and anti-metastasis effectof metformin. Mechanistically, AMPK phosphorylates PHF2 at S655 site, enhancing PHF2 demethylation activity and triggering thetranscription of CDH1. Furthermore, the PHF2-S655E mutant that mimics AMPK-mediated phosphorylation status further reducesH3K9me2 and suppresses lung cancer metastasis, while PHF2-S655A mutant presents opposite phenotype and reverses the antimetastasiseffect of metformin. PHF2-S655 phosphorylation strikingly reduces in lung cancer patients and the higherphosphorylation level predicts better survival.
