4Deficiency of mitochondrial outer membrane protein 64 confers rice resistance to both piercing-sucking and chewing insects in rice显示文摘The brown planthopper(BPH)and striped stem borer(SSB)are the most devastating insect pests in rice(Oryza sativa)producing areas.Screening for endogenous resistant genes is the most practical strategy for rice insect-resistance breeding.Forty-five mutants showing high re-sistance against BPH were identified in a rice T-DNA in-sertion population(11,000 putative homozygous lines)after 4 years of large-scale field BPH-resistance phenotype screening.Detailed analysis showed that deficiency of rice mitochondrial outer membrane protein 64(OM64)gene resulted in increased resistance to BPH.Mitochondrial outer membrane protein 64 protein is located in the outer mitochondrial membrane by subcellular localization and its deficiency constitutively activated hydrogen peroxide(H2O2)signaling,which stimulated antibiosis and tolerance to BPH.The om64 mutant also showed enhanced resist-ance to SSB,a chewing insect,which was due to promotion of Jasmonic acid biosynthesis and related responses.Im-portantly,om64 plants presented no significant changes in rice yield-related characters.This study confirmed OM64 as a negative regulator of rice herbivore resistance through regulating H2O2 production.Mitochondrial outer membrane protein 64 is a potentially efficient candidate to improve BPH and SSB resistance through gene deletion.Why the om64 mutant was resistant to both piercing-sucking and chewing insects via a gene deficiency in mitochondria is discussed.Hui‐Min Guo Hai‐Chao Li Shi‐Rong Zhou Hong‐Wei Xue Xue‐Xia Miao 2020Journal of Integrative Plant Biology2020,62,12:1

17Molecular and structural characterization of a promiscuous chalcone synthase from the fern species Stenoloma chusanum显示文摘The key enzymes involved in the flavonoid biosynthesis pathway have been extensively studied in seed plants,but relatively less in ferns.In this study,two 4-Coumarate:coenzyme A ligases(Sc4CL1 and Sc4CL2)and one novel chalcone synthase(ScCHS1)were functionally characterized by mining the Stenoloma chusanum transcriptome database.Recombinant Sc4CLs were able to esterify various hydroxycinnamic acids to corresponding acyl-coenzyme A(CoA).ScCHS1could catalyze p-coumaroyl-CoA,cinnamoyl-CoA,caffeoyl-CoA,and feruloyl-CoA to form naringenin,pinocembrin,eriodictyol,and homoeriodictyol,respectively.Moreover,enzymatic kinetics studies revealed that the optimal substrates of ScCHS1were feruloyl-CoA and caffeoyl-CoA,rather than p-coumaroyl-CoA,which was substantially different from the common CHSs.Crystallographic and sitedirected mutagenesis experiments indicated that the amino acid residues,Leu87,Leu97,Met165,and Ile200,located in the substrate-binding pocket near the B-ring of products,could exert a significant impact on the unique catalytic activity of ScCHS1.Furthermore,overexpression of ScCHS1 in tt4 mutants could partially rescue the mutant phenotypes.Finally,ScCHS1 and Sc4CL1 were used to synthesize flavanones and flavones with multi-substituted hydroxyl and methoxyl B-ring in Escherichia coli,which can effectively eliminate the need for the cytochrome P450 hydroxylation/O-methyltransferase from simple phenylpropanoid acids.In summary,the identification of these important Stenoloma enzymes provides a springboard for the future production of various flavonoids in E.coli.Rong Ni Meng Niu Jie Fu Hui Tan Ting‐Ting Zhu Jing Zhang Hong‐Xiang Lou Peng Zhang Jian‐Xu Li Ai‐Xia Cheng 2022Journal of Integrative Plant Biology2022,64,10:1
