1Different treatment strategies and molecular features between right-sided and left-sided colon cancers显示文摘The colon is derived from the embryological midgut and hindgut separately,with the right colon and left colon having different features with regards to both anatomical and physiological characteristics.Cancers located in the right and left colon are referred to as right colon cancer(RCC) and left colon cancer(LCC),respectively,based on their apparent anatomical positions.Increasing evidence supports the notion that not only are there differences in treatment strategies when dealing with RCC and LCC,but molecular features also vary between them,not to mention the distinguishing clinical manifestations.Disease-free survival after radical surgery of both RCC and LCC are similar.In the treatment of RCC,the benefit gained from adjuvant FOLFIRI chemotherapy is superior,or at least similar,to LCC,but inferior to LCC if FOLFOX regimen is applied.On the other hand,metastatic LCC exhibits longer survival than that of RCC in a palliative chemotherapy setting.For KRAS wild-type cancers,LCC benefits more from cetuximab treatment than RCC.Moreover,advanced LCC shows a higher sensitivity to bevacizumab treatment in comparison with advanced RCC.Significant varieties exist at the molecular level between RCC and LCC,which may serve as the cause of all apparent differences.With respect to carcinogenesis mechanisms,RCC is associated with known gene types,such as MMR,KRAS,BRAF,and mi RNA-31,while LCC is associated with CIN,p53,NRAS,mi RNA-146 a,mi RNA-147 b,and mi RNA-1288.Regarding protein expression,RCC is related to GNAS,NQO1,telomerase activity,P-PDH,and annexin A10,while LCC is related to Topo I,TS,and EGFR.In addition,separated pathways dominate progressionto relapse in RCC and LCC.Therefore,RCC and LCC should be regarded as two heterogeneous entities,with this heterogeneity being used to stratify patients in order for them to have the optimal,current,and novel therapeutic strategies in clinical practice.Additional research is needed to uncover further differences between RCC and LCC.Hong Shen Jiao Yang Qing Huang Meng-Jie Jiang Yi-Nuo Tan Jian-Fei Fu Li-Zhen Zhu Xue-Feng Fang Ying Yuan 2015World Journal of Gastroenterology2015,21,21:33
2EXPERIMENTAL STUDY OF FLOWFIELD STRUCTURE AROUND AN OGIVE-CYLINDER显示文摘The flowfield structure and their aerodynamic characteristics over an ogive cylinder were studied by means of flow visualization and surface pressure measurement in a water tunnel and a wind tunnel. The existence of multi asymmetric vortices over long slender bodies was experimentally confirmed at large angles of attack and in the subcritical Reynolds number range. The spatial 3 D characteristics of the multi vortices system were analyzed and a physical model was developed. The topological structure of different patterns in cross flow plane was studied and the mechanism governing the formation of asymmetric vortices and multi vortices was discussed from the viewpoint of stability of the topological structure. 3The China Multi-Model Ensemble Prediction System and Its Application to Flood-Season Prediction in 2018显示文摘Multi-model ensemble prediction is an effective approach for improving the prediction skill short-term climate prediction and evaluating related uncertainties. Based on a combination of localized operation outputs of Chinese climate models and imported forecast data of some international operational models, the National Climate Center of the China Meteorological Administration has established the China multi-model ensemble prediction system version 1.0 (CMMEv1.0) for monthly-seasonal prediction of primary climate variability modes and climate elements. We verified the real-time forecasts of CMMEv1.0 for the 2018 flood season (June-August) starting from March 2018 and evaluated the 1991-2016 hindcasts of CMMEv1.0. The results show that CMMEv1.0 has a significantly high prediction skill for global sea surface temperature (SST) anomalies, especially for the El Nino-Southern Oscillation (ENSO) in the tropical central-eastern Pacific. Additionally, its prediction skill for the North Atlantic SST triple (NAST) mode is high, but is relatively low for the Indian Ocean Dipole (IOD) mode. Moreover, CMMEv1.0 has high skills in predicting the western Pacific subtropical high (WPSH) and East Asian summer monsoon (EASM) in the June-July-August (JJA) season. The JJA air temperature in the CMMEv1.0 is predicted with a fairly high skill in most regions of China, while the JJA precipitation exhibits some skills only in northwestern and eastern China. For real-time forecasts in March-August 2018, CMMEv1.0 has accurately predicted the ENSO phase transition from cold to neutral in the tropical central-eastern Pacific and captures evolutions of the NAST and IOD indices in general. The system has also captured the main features of the summer WPSH and EASM indices in 2018, except that the predicted EASM is slightly weaker than the observed. 4Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 8Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 10Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 11Blockade of Endogenous Angiotensin-(1–7) in Hypothalamic Paraventricular Nucleus Attenuates High Salt-Induced Sympathoexcitation and Hypertension显示文摘Angiotensin(Ang)-(1–7) is an important biologically-active peptide of the renin-angiotensin system. This study was designed to determine whether inhibition of Ang-(1–7) in the hypothalamic paraventricular nucleus(PVN) attenuates sympathetic activity and elevates blood pressure by modulating pro-inflammatory cytokines(PICs)and oxidative stress in the PVN in salt-induced hypertension. Rats were fed either a high-salt(8% NaCl) or a normal salt diet(0.3% NaCl) for 10 weeks, followed by bilateral microinjections of the Ang-(1–7) antagonist A-779 or vehicle into the PVN. We found that the mean arterial pressure(MAP), renal sympathetic nerve activity(RSNA), and plasma norepinephrine(NE) were significantly increased in salt-induced hypertensive rats. The high-salt diet also resulted in higher levels of the PICs interleukin-6, interleukin-1 beta, tumor necrosis factor alpha, and monocyte chemotactic protein-1, as well as higher gp91 phoxexpression and superoxide production in the PVN. Microinjection of A-779(3 nmol/50 nL) into the bilateral PVN of hypertensive rats not only attenuated MAP, RSNA, and NE, but also decreased the PICs and oxidative stress in the PVN. 14The Xanthomonas typeⅢeffector Xop AP prevents stomatal closure by interfering with vacuolar acidification显示文摘Plant stomata close rapidly in response to a rise in the plant hormone abscisic acid(ABA)or salicylic acid(SA)and after recognition of pathogenassociated molecular patterns(PAMPs).Stomatal closure is the result of vacuolar convolution,ion efflux,and changes in turgor pressure in guard cells.Phytopathogenic bacteria secrete typeⅢeffectors(T3Es)that interfere with plant defense mechanisms,causing severe plant disease symptoms.Here,we show that the virulence and infection of Xanthomonas oryzae pv.oryzicola(Xoc),which is the causal agent of rice bacterial leaf streak disease,drastically increased in transgenic rice(Oryza sativa L.)plants overexpressing the Xoc T3E gene Xop AP,which encodes a protein annotated as a lipase.We discovered that Xop AP binds to phosphatidylinositol 3,5-bisphosphate(Ptd Ins(3,5)P_(2)),a memb rane phospholipid that functions in p H control in lysosomes,membrane dynamics,and protein trafficking.Xop AP inhibited the acidification of vacuoles by competing with vacuolar H^(+)-pyrophosphatase(V-PPase)for binding to Ptd Ins(3,5)P_(2),leading to stomatal opening.Transgenic rice overexpressing Xop AP also showed inhibition of stomatal closure when challenged by Xoc infection and treatment with the PAMP flg22.Moreover,Xop AP suppressed flg22-induced gene expression,reactive oxygen species burst and callose deposition in host plants,demonstrating that Xop AP subverts PAMP-triggered immunity during Xoc infection.Taken together,these findings demonstrate that Xop AP overcomes stomatal immunity in plants by binding to lipids.Longyu Liu Ying Li Zhengyin Xu Huan Chen Jingyi Zhang Brittany Manion Fengquan Liu Lifang Zou Zheng Qing Fu Gongyou Chen 2022Journal of Integrative Plant Biology2022,64,10:3
15Fabrication and Evaluation of Porous Keratin/chitosan(KCS) Scaffolds for Effectively Accelerating Wound Healing显示文摘Objective To develop a dressing with desired antibacterial activity, good water maintaining ability and mechanical properties for wound healing and skin regeneration. Methods The chitosan with different concentrations were added in keratin solution to form porous keratin/chitosan(KCS) scaffolds. The morphological characteristics, chemical composition, wettability, porosity, swelling ratio and degradation of the scaffolds were evaluated. The antibacterial activity was tested by using S. aureus and E. coli suspension for 2 h. And L929 fibroblast cells culture was used to evaluate the cytotoxicity of the KCS scaffolds. Results The adding of chitosan could increase the hydrophobicity, decrease porosity, swelling ratio and degradation rate of the KCS porous scaffolds. Mechanical properties of KCS scaffolds could be enhanced and well adjusted by chitosan. KCS scaffolds could obviously decrease bacteria number. The proliferation of fibroblast cells in porous KCS patch increased firstly and then decreased with the increase of chitosan concentration. It was appropriate to add 400 μg/m L chitosan to form porous KCS scaffold for achieving best cell attachment and proliferation compared with other samples.
