2Inhibitory effects of saikosaponin-d on CCl_4-induced hepatic fibrogenesis in rats显示文摘AIM: To investigate the suppressive effect of saikosaponin-d (SSd) on hepatic fibrosis in rats induced by CCl4 injections in combination with alcohol and high fat, low protein feeding and its relationship with the expression of nuclear factor-κB (NF-κB), tumor necrosis factor-alpha (TNF-α) and interleukins-6 (IL-6). METHODS: Hepatic fibrosis models were induced by subcutaneous injection of CCl4 at a dosage of 3 mL/kg in rats. At the same time, rats in treatment groups were injected intraperitoneally with SSd at different doses (1.0, 1.5 and 2.0 mg/kg) once daily for 6 wk in combination with CCl4, while the control group received olive oil instead of CCl4. At the end of the experiment, rats were anesthetized and killed (except for 8 rats which died during the experiment; 2 from the model group, 3 in high-dose group, 1 in medium-dose group and 2 in low- dose group). Hematoxylin and eosin (HE) staining and Van Gieson staining were used to examine the changes in liver pathology. The levels of alanine aminotransferase (ALT), triglyeride (TG), albumin (ALB), globulin (GLB), hyaluronic acid (HA) and laminin (LN) in serum and the content of hydroxyproline (HYP) in liver were measured by biochemical examinations and radioimmuneoassay, respectively. In addition, the expression of TNF-α and IL-6 in liver homogenate was evaluated by enzyme- linked immunosorbent assay (ELISA) and the levels of NF-κBp65 and I-κBα in liver tissue were analyzed by Western blotting. RESULTS: Both histological examination and Van Gieson staining demonstrated that SSd could attenuate the area and extent of necrosis and reduce the scores of liver fibrosis. Similarly, the levels of ALT, TG, GLB, HA, andLN in serum, and the contents of HYP, TNF-α and IL-6 in liver were all significantly increased in model group in comparison with those in control group. Whereas, the treatment with SSd markedly reduced all the above parameters compared with the model group, especially in the medium group (ALT: 412 ± 94.5 IU/L vs 113.76 ± 14.91 IU/L, TG: 0.95 ± 0.16 mmol/L vs 0.51 ± 0.06 mmol/L, GLB: 35.62 ± 3.28 g/L vs 24.82 ± 2.73 g/L, HA: 42.15 ± 8.25 ng/mL vs 19.83 ± 3.12 ng/mL, LN: 27.56 ± 4.21 ng/mL vs 13.78 ± 2.57 ng/mL, HYP: 27.32 ± 4.32 μg/mg vs 16.20 ± 3.12 μg/mg, TNF-α: 4.38 ± 0.76 ng/L vs 1.94 ± 0.27 ng/L, IL-6: 28.24 ± 6.37 pg/g vs 12.72 ± 5.26 pg/g, respectively, P < 0.01). SSd also decreased ALB in serum (28.49 ± 4.93 g/L vs 37.51 ± 3.17 g/L, P < 0.05). Moreover, the expression of NF-κB p65 in the liver of treated groups was lower than that in model groups while the expression of I-κBα was higher in treated group than in model group (P < 0.01). The expression of NF-κBp65 and TNF-α had a positive correlation with the level of HA in serum of rats after treatment with CCl4 (r = 0.862, P < 0.01; r = 0.928, P < 0.01, respectively). 5Add-on pegylated interferon augments hepatitis B surface antigen clearance vs continuous nucleos(t)ide analog monotherapy in Chinese patients with chronic hepatitis B and hepatitis B surface antigen≤1500 IU/mL:An observational study显示文摘BACKGROUND Nucleos(t)ide analog(NA)has shown limited effectiveness against hepatitis B surface antigen(HBsAg)clearance in chronic hepatitis B(CHB)patients.AIM To evaluate the efficacy and safety of add-on peginterferonα-2a(peg-IFNα-2a)to an ongoing NA regimen in CHB patients.METHODS In this observational study,195 CHB patients with HBsAg≤1500 IU/m L,hepatitis B e antigen(HBeAg)-negative(including HBeAg-negative patients or HBeAg-positive patients who achieved HBeAg-negative after antiviral treatment with NA)and hepatitis B virus-deoxyribonucleic acid<1.0×10^2 IU/mL after over 1 year of NA therapy were enrolled between November 2015 and December2018 at the Second Affiliated Hospital of Xi'an Jiaotong University,China.Patients were given the choice between receiving either peg-IFNα-2a add-on therapy to an ongoing NA regimen(add-on group,n=91)or continuous NA monotherapy(monotherapy group,n=104)after being informed of the benefits and risks of the peg-IFNα-2a therapy.Total therapy duration of peg-IFNα-2a was 48 wk.All patients were followed-up to week 72(24 wk after discontinuation of peg-IFNα-2a).The primary endpoint was the proportion of patients with HBsAg clearance at week 72.RESULTS Demographic and baseline characteristics were comparable between the two groups.Intention-to-treatment analysis showed that the HBsAg clearance rate in the add-on group and monotherapy group was 37.4%(34/91)and 1.9%(2/104)at week 72,respectively.The HBsAg seroconversion rate in the add-on group was 29.7%(27/91)at week 72,and no patient in the monotherapy group achieved HBsAg seroconversion at week 72.The HBsAg clearance and seroconversion rates in the add-on group were significantly higher than in the monotherapy group at week 72(P<0.001).Younger patients,lower baseline HBsAg concentration,lower HBsAg concentrations at weeks 12 and 24,greater HBsAg decline from baseline to weeks 12 and 24 and the alanine aminotransferase≥2×upper limit of normal during the first 12 wk of therapy were strong predictors of HBsAg clearance in patients with peg-IFNα-2a add-on treatment.Regarding the safety of the treatment,4.4%(4/91)of patients in the add-on group discontinued peg-IFNα-2a due to adverse events.No severe adverse events were noted.CONCLUSION Peg-IFNα-2a as an add-on therapy augments HBsAg clearance in HBeAg-negative CHB patients with HBsAg≤1500 IU/m L after over 1 year of NA therapy.Feng-Ping Wu Ying Yang Mei Li Yi-Xin Liu Ya-Ping Li Wen-Jun Wang Juan-Juan Shi Xin Zhang Xiao-Li Jia Shuang-Suo Dang 2020World Journal of Gastroenterology2020,26,13:27

7Inhibitory effect of emodin and Astragalus polysaccharide on the replication of HBV显示文摘AIM: To evaluate the anti-viral effect of emodin plus Astragalus polysaccharide (APS) in hepatitis B virus (HBV) transgenic mice.METHODS: Sixty HBV transgenic mice (HBV TGM) whose weight varied between 18 and 24 g were randomly divided into 3 groups, with 20 mice in each group. Group A was the normal control, where the mice were treated with physiological saline; group B was the positive control where the mice were treated with lamivudine solution (100 mL/kg per day). Group C was the experimental group where the mice were treated with physiological saline containing emodin and APS (57.59 mg/kg per day and 287.95 mg/kg per day, respectively). The mice were treated daily for 3 wk. After 1 wk recovery time, the mice were sacrifi ced and serum as well as liver tissues were collected for ELISA and histological examination.RESULTS: After 21 d treatment, HBV DNA levels in group B and group C significantly declined when compared with group A (P < 0.05). However, a signif icant increase in HBV DNA content was observed in group B, whereas this phenomenon was not observed in group C. 8Surveillance-based evidence:elimination of schistosomiasis as a public health problem in the Peoples’Republic of China显示文摘Background:A steady progress on schistosomiasis control in the Peoples’Republic of China(P.R.China)was achieved and broadened into the twelve-year medium and long term national plan(MLNP)which marled the implementation of an integrated control strategy across all endemic areas in P.R.China in 2004.To understand the endemic trends of schistosomiasis to assess the effectiveness of an integrated strategy,we conducted an analysis of schistosomiasis surveillance data spanned from 2005 to 2015.Methods:The schistosomiasis sentinel surveillance data from sentinel sites were collected and analyzed from 2005 to 2015.In these sentinel sites,residents aged 6 years or above were screened annually by indirect hemagglutination assay(IHA),while only antibody positives were followed by stool examination either Kato-katz method(KK)and/or hatching technique(HT).Domestic animals raised in sentinel sites were examined by HT for confirming the infection of schistosomes.Snail investigation was conducted each year through systematic sampling method combined with environmental sampling method.The snails collected from field were tested by microscopic dissection method.The infection rates of schistosomes in residents,domestic animals and snails,as well as the indicators reflecting the snails’distribution were calculated and analyzed.ANOVA analysis was used to examine the changes of the number of eggs per gram feces in population and Chi-square test was used to examine any change in proportions among groups.Results:A total of 148902 residents from sentinel sites attended this study and 631676 blood samples were examined by IHA test during the 11 covered years.The annual average antibody positive rates presented a significant decrease trends,from 17.48%(95%CI:17.20–17.75%)in 2005 to 5.93%(95%CI:5.71–6.15%)(χ2=8890.47,P<0.001)in 2015.During 2005–2015,the average infection rate of schistosomes in residents declined from 2.07%(95%CI:1.96–2.17%)to 0.13%(95%CI:0.09–0.16%),accompanied by significant decrease of infection intensity in population.In 2015,the stool positives were only found in farmers,fishermen and boatmen with infection rate of 0.16%(95%CI:0.11–0.20%),0.17%(95%CI:0–0.50%)respectively.The infection rate of schistosomes in domestic animals dropped from 9.42%(538/5711,95%CI:8.66–10.18%)to 0.08%(2/2360,95%CI:0–0.20%)from 2005 to 2015.Infections were found in eight species of domestic animals at the beginning of surveillance while only two cattle were infected in 2015.Totally 98 ha of new snail habitats were found,while 94.90%(93/98)distributed in lake and marshland regions.The percentage of frames with snails decreased from 16.96%(56884/335391,95%CI:16.83–17.09%)in 2005 to 4.28%(18121/423755,95%CI:4.22–4.34%)in 2014,with a slightly increase in 2015.Meanwhile,the infection rate of schistosomes in snails was decreased from 0.26%(663/256531,95%CI:0.24–0.28%)to zero during 2005–2015.Conclusions:The infection rate of schistosomes declined significantly,providing evidence that the goal of the MLNP was achieved.Elimination of schistosomiasis as a public health problem defined as WHO was also reached in P.R.China nationwide.Surveillance-response system should be improved and strengthened to realize the final goal of schistosomiasis elimination.Jing Xu Shi-Zhu Li Li-Juan Zhang Robert Bergquist Hui Dang Qiang Wang Shan Lv Tian-Ping Wang Dan-Dan Lin Jian-Bing Liu Guang-Hui Ren Kun Yang Yang Liu Yi Dong Shi-Qing Zhang Xiao-Nong Zhou 2020Infectious Diseases of Poverty2020,9,3:20

10Long noncoding RNAs in hepatocellular carcinoma: Novel insights into their mechanism显示文摘Hepatocellular carcinoma(HCC) is the predominant subject of liver malignancies which arouse global concern. Advanced studies have found that long noncoding RNAs(lnc RNAs) are differentially expressed in HCC and implicate they may play distinct roles in the pathogenesis and metastasis of HCC. However, the underlying mechanisms remain largely unclear. In this review, we summarized the functions and mechanisms of those known aberrantly expressed lncR NAs identified in human HCC tissues. We hope to enlighten more comprehensive researches on the detailed mechanisms of lncR NAs and their application in clinic, such as being used as diagnostic and prognostic biomarkers and the targets for potential therapy. 11Inorganic Phosphorus Fractions and Phosphorus Availability in a Calcareous Soil Receiving 21-Year Superphosphate Application显示文摘A long-term (21-year) field experiment was performed to study the responses of soil inorganic P fractions and P availability to annual fertilizer P application in a calcareous soil on the Loess Plateau of China. Soil Olsen-P contents increased by 3.7, 5.2, 11.2 and 20.6 mg P kg-1 after 21-year annual fertilizer P application at 20, 39, 59, and 79 kg P ha-1, respectively. Long-term fertilizer P addition also increased soil total P and inorganic P (Pi) contents significantly. The contents of inorganic P fractions were in the order of Ca10-P > Ca8-P > Fe-P > Al-P > occluded P > Ca2-P in the soil receiving annual fertilizer P application. Fertilizer P application increased Ca8-P, Al-P and Ca2-P contents as well as their percentages relative to Pi. Pi application increased Fe-P and occluded P contents but nor their percentages. Soil Ca10-P content remained unchanged after fertilizer P application while its percentage relative to Pi declined with increasing fertilizer P rate. All Pi fractions but Ca10-P were correlated with Olsen-P significantly. 90% of variations in Olsen-P could be explained by Pi fractions, and the direct contribution of Ca8-P was predominant. 13Undersea safety mining of the large gold deposit in Xinli District of Sanshandao Gold Mine显示文摘The exploration of undersea resources becomes popular as land resources decrease. Researches were conducted with emphasis on the safety and efficiency of undersea mining of the large gold deposit in Xinli District of Sanshandao Gold Mine. A series of tests for the physical and mechanical characteristics of rock mass were carried out, and the three-dimensional geo-stress distribution was tested in the mining area. Further, a similar experimental simulation platform, which revealed the mechanism of water inrush and ascertained the reasonable thickness of the safety isolate layer, was established for the undersea mining. Meanwhile, the feasibility of cancelling the ore pillars and the safety conditions was checked by numerical simulation. The simulation results show that it is safe to exploit the ore body below the 85 m level (presently, the exploitation level is below 160 m in Xinli District), and the ore pillars can be cancelled below the 560 m level. Furthermore, a novel backfill method was designed to reduce the rock strata disturbance and settlement, and the settlement of roof strata was monitored during the mining process. Engineering practice shows that the settlement of roof strata was small and that no disaster happened. 14Investigation of gas content of organic-rich shale:A case study from Lower Permian shale in southern North China Basin,central China显示文摘Measuring gas content is an essential step in estimating the commerciality of gas reserves. In this study,eight shale core samples from the Mouye-1 well were measured using a homemade patented gas desorption apparatus to determine their gas contents. Due to the air contamination that is introduced into the desorption canister, a mathematical method was devised to correct the gas quantity and quality.Compared to the chemical compositions of desorbed gas, the chemical compositions of residual gas are somewhat different. In residual gas, carbon dioxide and nitrogen record a slight increase, and propane is first observed. This phenomenon may be related to the exposure time during the transportation of shale samples from the drilling site to the laboratory, as well as the differences in the mass, size and adsorptivity of different gas molecules. In addition to a series of conventional methods, including the USBM direct method and the Amoco Curve Fit(ACF) method, which were used here for lost gas content estimation, a Modified Curve Fit(MCF) method, based on the 'bidisperse' diffusion model, was established to estimate lost gas content. By fitting the ACF and MCF models to gas desorption data, we determined that the MCF method could reasonably describe the gas desorption data over the entire time period, whereas the ACF method failed. The failure of the ACF method to describe the gas desorption process may be related to its restrictive assumption of a single pore size within shale samples. In comparison to the indirect method, this study demonstrates that none of the three methods studied in this investigation(USBM, ACF and MCF) could individually estimate the lost gas contents of all shale samples and that the proportion of free gas relative to total gas has a significant effect on the estimation accuracy of the selected method. 18Carbon pools and fluxes in the China Seas and adjacent oceans显示文摘The China Seas include the South China Sea, East China Sea, Yellow Sea, and Bohai Sea. Located off the Northwestern Pacific margin, covering 4700000 km^2 from tropical to northern temperate zones, and including a variety of continental margins/basins and depths, the China Seas provide typical cases for carbon budget studies. The South China Sea being a deep basin and part of the Western Pacific Warm Pool is characterized by oceanic features; the East China Sea with a wide continental shelf, enormous terrestrial discharges and open margins to the West Pacific, is featured by strong cross-shelf materials transport; the Yellow Sea is featured by the confluence of cold and warm waters; and the Bohai Sea is a shallow semiclosed gulf with strong impacts of human activities. Three large rivers, the Yangtze River, Yellow River, and Pearl River, flow into the East China Sea, the Bohai Sea, and the South China Sea, respectively. The Kuroshio Current at the outer margin of the Chinese continental shelf is one of the two major western boundary currents of the world oceans and its strength and position directly affect the regional climate of China. These characteristics make the China Seas a typical case of marginal seas to study carbon storage and fluxes. This paper systematically analyzes the literature data on the carbon pools and fluxes of the Bohai Sea,Yellow Sea, East China Sea, and South China Sea, including different interfaces(land-sea, sea-air, sediment-water, and marginal sea-open ocean) and different ecosystems(mangroves, wetland, seagrass beds, macroalgae mariculture, coral reefs, euphotic zones, and water column). Among the four seas, the Bohai Sea and South China Sea are acting as CO_2 sources, releasing about0.22 and 13.86–33.60 Tg C yr^(-1) into the atmosphere, respectively, whereas the Yellow Sea and East China Sea are acting as carbon sinks, absorbing about 1.15 and 6.92–23.30 Tg C yr^(-1) of atmospheric CO_2, respectively. Overall, if only the CO_2 exchange at the sea-air interface is considered, the Chinese marginal seas appear to be a source of atmospheric CO_2, with a net release of 6.01–9.33 Tg C yr^(-1), mainly from the inputs of rivers and adjacent oceans. The riverine dissolved inorganic carbon (DIC) input into the Bohai Sea and Yellow Sea, East China Sea, and South China Sea are 5.04, 14.60, and 40.14 Tg C yr^(-1),respectively. The DIC input from adjacent oceans is as high as 144.81 Tg C yr^(-1), significantly exceeding the carbon released from the seas to the atmosphere. In terms of output, the depositional fluxes of organic carbon in the Bohai Sea, Yellow Sea, East China Sea, and South China Sea are 2.00, 3.60, 7.40, and 5.92 Tg C yr^(-1), respectively. The fluxes of organic carbon from the East China Sea and South China Sea to the adjacent oceans are 15.25–36.70 and 43.93 Tg C yr^(-1), respectively. The annual carbon storage of mangroves, wetlands, and seagrass in Chinese coastal waters is 0.36–1.75 Tg C yr^(-1), with a dissolved organic carbon(DOC) output from seagrass beds of up to 0.59 Tg C yr^(-1). Removable organic carbon flux by Chinese macroalgae mariculture account for 0.68 Tg C yr^(-1) and the associated POC depositional and DOC releasing fluxes are 0.14 and 0.82 Tg C yr^(-1), respectively. Thus, in total, the annual output of organic carbon, which is mainly DOC, in the China Seas is 81.72–104.56 Tg C yr^(-1). The DOC efflux from the East China Sea to the adjacent oceans is 15.00–35.00 Tg C yr^(-1). The DOC efflux from the South China Sea is 31.39 Tg C yr^(-1). 19MiRNA-429 suppresses the growth of gastric cancer cells in vitro显示文摘Micro-RNAs(miRNAs) have been found to be implicated in a very wide range of physiological processes.This study was aimed to investigate the regulation of miRNA-429(miR-429) in gastric cancer cells on cell proliferation and apoptosis.Quantitative PCR was employed to detect the expressions of miR-429 after eukaryotic expression plasmid of miR-429 and its inhibitor were transiently transfected into poorly differentiated human gastric can-cer cell line BGC823.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) reduction as-says were used to examine proliferation ability.Apoptosis was analyzed by flow cytometry after transfection.The results showed that 48 h after transfection,overexpression of miR-429 reached maximum efficiency.Compared with mock transfection,miR-429 inhibited tumor cell proliferation significantly(P < 0.05) at 48 h and 72 h.of Overexpression of miR-429 promoted tumor cell apoptosis when compared with mock transfected cells(P < 0.05).On the contrary,miR-429 inhibitor promoted tumor cell proliferation and inhibited apoptosis when compared with controls(P < 0.05).Our results suggested that miRNA-429 may serve as a tumor suppressor during tumorigenesis of gastric cancer and may be a potential gastric cancer therapeutic target.Di Liu Peng Xia Dongmei Diao Yao Cheng Hao Zhang Dawei Yuan Chen Huang Chengxue Dang 2012The Journal of Biomedical Research2012,26,5:12
20New strategy for designing orangish-redemitting phosphor via oxygen-vacancyinduced electronic localization显示文摘Phosphor-converted white-light-emitting diodes(pc-WLED)have been extensively employed as solid-state lighting sources,which have a very important role in people’s daily lives.However,due to the scarcity of the red component,it is difficult to realize warm white light efficiently.Hence,red-emitting phosphors are urgently required for improving the illumination quality.In this work,we develop a novel orangish-red La_(4)GeO_(8):Bi^(3+) phosphor,the emission peak of which is located at 600 nm under near-ultraviolet(n-UV)light excitation.The full width at half maximum(fwhm)is 103 nm,the internal quantum efficiency(IQE)exceeds 88%,and the external quantum efficiency(EQE)is 69%.According to Rietveld refinement analysis and density functional theory(DFT)calculations,Bi^(3+) ions randomly occupy all La sites in orthorhombic La_(4)GeO_(8).Importantly,the oxygen-vacancy-induced electronic localization around the Bi3+ions is the main reason for the highly efficient orangish-red luminescence.These results provide a new perspective and insight from the local electron structure for designing inorganic phosphor materials that realize the unique luminescence performance of Bi^(3+) ions.Yi Wei Gongcheng Xing Kang Liu Guogang Li Peipei Dang Sisi Liang Min Liu Ziyong Cheng Dayong Jin Jun Lin 2019Light(Science & Applications)2019,8,1:12
