3Abscisic Acid Connects Phytohormone Signaling with RNA Metabolic Pathways and Promotes an Antiviral Response that Is Evaded by a Self- Controlled RNA Virus显示文摘A complex network of cellular receptors,RNA targeting pathways, and small-molecule signaling provides robust plant immunity and tolerance to viruses. To maximize their fitness, viruses must evolve control mechanisms to balance host immune evasion and plant-damaging effects. The genus Potyvirus comprises plant viruses characterized by RNA genomes that encode large polyproteins led by the P1 protease.A P1 autoinhibitory domain controls polyprotein processing, the release of a downstream functional RNAsilencing suppressor, and viral replication. Here,we show that P1Pro,a plum pox virus clone that lacks the P1 autoinhibitory domain, triggers complex reprogramming of the host transcriptome and high levels of abscisic acid(ABA)accumulation.A meta-analysis highlighted ABA connections with host pathways known to control RNA stability, turnover, maturation, and translation.Transcriptomic changes triggered by P1Pro infection or ABA showed similarities in host RNA abundance and diversity.Genetic and hormone treatment assays showed that ABA promotes plant resistance to potyviral infection. Finally, quantitative mathematical modeling of viral replication in the presence of defense pathways supported self-control of polyprotein processing kinetics as a viral mechanism that attenuates the magnitude of the host antiviral response.Overall, our findings indicate that ABA is an active player in plant antiviral immunity, which is nonetheless evaded by a self-controlled RNA virus.Fabio Pasin Hongying Shan Beatriz García Maren Müller David San León Márta Ludman David H.Fresno Károly Fátyol Sergi Munné-Bosch Guillermo Rodrigo Juan Antonio García 2020Plant Communications2020,1,5:1

8Host-parasite interactions in perpetual darkness:Macroparasite diversity in the cavefish Astyanax mexicanus显示文摘Astyanax mexicanus has repeatedly colonized cave environments, displaying evolutionary parallelisms in many troglobitic traits.Despite being a model system for the study of adaptation to life in perpetual darkness, the parasites that infect cavefish are practically unknown. In this study,we investigated the macroparasite communities in 18 cavefish populations from independent lineages and compared them with the parasite diversity found in their sister surface fish populations, with the aim of better understanding the role that parasites play in the colonization of new environments.Within the cavefish populations,we identified 13 parasite taxa, including a subset of 10 of the 27 parasite taxa known for the surface populations.Parasites infecting the cavefish belong to five taxonomic groups, including trematodes, monogeneans, nematodes, copepods, and acari.Monogeneans are the most dominant group, found in 14 caves. The macroparasites include species with direct life cycles and trophic transmission, including invasive species.Surprisingly, paired comparisons indicate higher parasite richness in the cavefish than in the surface fish.Spatial variation in parasite composition across the caves suggests historical and geographical contingencies in the host-parasite colonization process and potential evolution of local adaptations. This base-line data on parasite diversity in cavefish populations of A.mexicanus provides a foundation to explore the role of divergent parasite infections under contrasting ecological pressures(cave vs.surface environments)in the evolution of cave adaptive traits.Ana Santacruz David Hernández-Mena Ramses Miranda-Gamboa Gerardo Pérez-Ponce De León Claudia Patricia Ornelas-García 2023Zoological Research2023,44,4:0
9Decline in subarachnoid haemorrhage volumes associated with the first wave of the COVID- 19 pandemic显示文摘Background During the COVID-19 pandemic, decreased volumes of stroke admissions and mechanical thrombectomy were reported. The study’s objective was to examine whether subarachnoid haemorrhage(SAH)hospitalisations and ruptured aneurysm coiling interventions demonstrated similar declines. Methods We conducted a cross-sectional,retrospective,observational study across 6 continents,37 countries and 140 comprehensive stroke centres.Patients with the diagnosis of SAH,aneurysmal SAH,ruptured aneurysm coiling interventions and COVID-19 were identified by prospective aneurysm databases or by International Classification of Diseases,10th Revision, codes. The 3-month cumulative volume, monthly volumes for SAH hospitalisations and ruptured aneurysm coiling procedures were compared for the period before(1 year and immediately before)and during the pandemic, defined as 1 March-31 May 2020.The prior 1-year control period(1 March-31 May 2019)was obtained to account for seasonal variation.Findings There was a significant decline in SAH hospitalisations, with 2044 admissions in the 3 months immediately before and 1585 admissions during the pandemic, representing a relative decline of 22.5%(95%CI−24.3%to−20.7%,p<0.0001).Embolisation of ruptured aneurysms declined with 1170-1035 procedures, respectively, representing an 11.5%(95%CI−13.5%to−9.8%,p=0.002)relative drop.Subgroup analysis was noted for aneurysmal SAH hospitalisation decline from 834 to 626 hospitalisations,a 24.9%relative decline(95%CI−28.0%to−22.1%,p<0.0001).A relative increase in ruptured aneurysm coiling was noted in low coiling volume hospitals of 41.1%(95%CI 32.3%to 50.6%,p=0.008)despite a decrease in SAH admissions in this tertile.Interpretation There was a relative decrease in the volume of SAH hospitalisations, aneurysmal SAH hospitalisations and ruptured aneurysm embolisations during the COVID-19 pandemic. These findings in SAH are consistent with a decrease in other emergencies, such as stroke and myocardial infarction.Thanh N Nguyen Diogo C Haussen Muhammad M Qureshi Hiroshi Yamagami 10Ventilation practices in burn patients—an international prospective observational cohort study显示文摘Background:It is unknown whether lung-protective ventilation is applied in burn patients and whether they benefit from it.This study aimed to determine ventilation practices in burn intensive care units(ICUs)and investigate the association between lung-protective ventilation and the number of ventilator-free days and alive at day 28(VFD-28).Methods:This is an international prospective observational cohort study including adult burn patients requiring mechanical ventilation.Low tidal volume(V_(T))was defined as V_(T)≤8 mL/kg predicted body weight(PBW).Levels of positive end-expiratory pressure(PEEP)and maximum airway pressures were collected. The association between V_(T) and VFD-28 was analyzed using a competing risk model.Ventilation settings were presented for all patients, focusing on the first day of ventilation. We also compared ventilation settings between patients with and without inhalation trauma. Results:A total of 160 patients from 28 ICUs in 16 countries were included.Low V_(T) was used in 74%of patients, median V_(T) size was 7.3[interquartile range(IQR)6.2–8.3]mL/kg PBW and did not differ between patients with and without inhalation trauma(p=0.58).Median VFD-28 was 17(IQR 0–26),without a difference between ventilation with low or high V_(T)(p=0.98).All patients were ventilated with PEEP levels≥5 cmH_(2)O;80%of patients had maximum airway pressures<30 cmH_(2)O.Conclusion:In this international cohort study we found that lung-protective ventilation is used in the majority of burn patients, irrespective of the presence of inhalation trauma.Use of low V_(T) was not associated with a reduction in VFD-28.Trial registration:Clinicaltrials.gov NCT02312869.Date of registration:9 December 2014.Gerie J.Glas Janneke Horn Markus W.Hollmann Benedikt Preckel Kirsten Colpaert Manu Malbrain Ary Serpa Neto Karim Asehnoune Marcello Gamma de Abreu Ignacio Martin-Loeches Paolo Pelosi Folke Sjoerg Jan M.Binnekade Berry Cleffken Nicole P.Juffermans Paul Knape Bert G.Loef David P.Mackie Perenlei Enkhbaatar Nadia Depetris Anders Perner Eva Herrero Lucia Cachafeiro Marc Jeschke Jeffrey Lipman Matthieu Legrand Johannes Horter Athina Lavrentieva Alex Kazemi Anne Berit Guttormsen Frederik Huss Mark Kol Helen Wong Therese Starr Luc De Crop Wilson de Oliveira Filho Joa Manoel Silva Junior Cintia M.C.Grion Marjorie Burnett Frederik Mondrup Francois Ravat Mathieu Fontaine Renan Le Floch Mathieu Jeanne Morgane Bacus Maite Chaussard Marcus Lehnhardt Bassem Daniel Mikhail Jochen Gille Aidan Sharkey Nicole Trommel Auke C.Reidinga Nadine Vieleers Anna Tilsley Henning Onarheim Maria Teresa Bouza Alexander Agrifoglio Filip Fredén Tina Palmieri Lynda E.Painting Marcus J.Schultz LAMiNAR investigators 2021Burns & Trauma2021,9,1:0
