| 8 | Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. | 11 | Sutureless Intestinal Anastomosis with a Novel Device of Magnetic Compression Anastomosis显示文摘Objective To explore the feasibility and efficiency of a novel magnetic compression anastomats(MCAs) in intestinal anastomosis.Methods A total of 36 male mongrel canines underwent intestinal anastomosis using traditional hand-sewn(n=18) or a novel MCAs(n=18).We compared the anastomosis time,postoperative complications,bursting strength of anastomoses,gross appearance,and pathology between two groups at each time-point of follow-up.Results The mean anastomosis time with MCAs was significantly less than that with hand-sewn(8.50±1.95 vs.31.1±4.32 minutes,P<0.001).The blood stools and intussusceptions occurred in both groups during follow-up period.Only 1 mongrel canine receiving intestinal anastomosis by MCAs experienced anastomotic leakage.The average bursting pressure of anastomoses obtained from mongrel canines undergoing intestinal anastomosis by MCAs was significantly higher than that by traditional hand-sewn at 1 week's follow-up time(P<0.05).Gross appearance of the anastomoses constructed by MCAs was relatively smoother and flatter.Pathological evalution of anastomoses revealed that general inflammation was greater in hand-sewn anastomoses than magnetic anastomosis.Conclusion The magnetic compression anastomat is a safe and effective device of sutureless intestinal anastomosis in canine models. | Chao Fan Jia Ma Hong-ke Zhang Rui Gao Jian-hui Li Liang Yu Zheng Wu Yi Lv | 2011 | Chinese Medical Sciences Journal2011,26,3: | 20 |

| 13 | A hybrid dynamic programming-rule based algorithm for real-time energy optimization of plug-in hybrid electric bus显示文摘The optimization of the control strategy of a plug-in hybrid electric bus(PHEB) for the repeatedly driven bus route is a key technique to improve the fuel economy. The widely used rule-based(RB) control strategy is lacking in the global optimization property, while the global optimization algorithms have an unacceptable computation complexity for real-time application. Therefore, a novel hybrid dynamic programming-rule based(DPRB) algorithm is brought forward to solve the global energy optimization problem in a real-time controller of PHEB. Firstly, a control grid is built up for a given typical city bus route, according to the station locations and discrete levels of battery state of charge(SOC). Moreover, the decision variables for the energy optimization at each point of the control grid might be deduced from an off-line dynamic programming(DP) with the historical running information of the driving cycle. Meanwhile, the genetic algorithm(GA) is adopted to replace the quantization process of DP permissible control set to reduce the computation burden. Secondly, with the optimized decision variables as control parameters according to the position and battery SOC of a PHEB, a RB control is used as an implementable controller for the energy management. Simulation results demonstrate that the proposed DPRB might distribute electric energy more reasonably throughout the bus route, compared with the optimized RB. | 14 | Preliminary results of environmental monitoring of the natural gas hydrate production test in the South China Sea显示文摘Natural gas hydrate (NGH)is considered as one of the new clean energy sources of the 21st century with the highest potential.The environmental issues of NGH production have attracted the close attention of scientists in various countries.From May 10 to July 9,2017,the first offshore NGH production test in the South China Sea (SCS)was conducted by the China Geological Survey.In addition,environmental security has also been effectively guaranteed via a comprehensive environmental monitoring system built during the NGH production test.The monitoring system considered sea-surface atmosphere methane and carbon dioxide concentrations,dissolved methane in the sea water column,and the seafloor physical oceanography and marine chemistry environment.The whole process was monitored via multiple means, in multiple layers,in all domains,and in real time.After the production test,an environmental investigation was promptly conducted to evaluate the environmental impact of the NGH production test. | 16 | BMAL1 regulates mitochondrial fission and mitophagy through mitochondrial protein BNIP3 and is critical in the development of dilated cardiomyopathy显示文摘Dysregulation of circadian rhythms associates with cardiovascular disorders.It is known that deletion of the core circadian gene Bma/1 in mice causes dilated car-diomyopathy.However,the biological rhythm regulation system in mouse is very different from that of humans.Whether BMAL1 plays a role in regulating human heart function remains unclear.Here we generated a BMAL1 knockout human embryonic stem cell(hESC)model and further derived human BMAL1 deficient cardiomy-ocytes.We show that BMAL1 deficient hESC-derived cardiomyocytes exhibited typical phenotypes of dilated cardiomyopathy including attenuated contractility,cal-cium dysregulation,and disorganized myofilaments.In addition,mitochondrial fission and mitophagy were suppressed in BMAL1 deficient hESC-cardiomyocytes,which resulted in significantly attenuated mitochondrial oxidative phosphorylation and compromised cardiomy-ocyte function.We also found that BMAL1 binds to the E-box element in the promoter region of BNIP3 gene and specifically controls BNIP3 protein expression.BMAL1 knockout directly reduced BNIP3 protein level,causing compromised mitophagy and mitochondria dysfunction and thereby leading to compromised cardiomyocyte function.Our data indicated that the core circadian gene S/VMLf is critical for normal mitochondria activities and cardiac function.Circadian rhythm disruption may directly link to compromised heart function and dilated cardiomyopathy in humans. | Ermin Li Xiuya Li Jie Huang Chen Xu Qianqian Liang Kehan Ren Aobing Bai Chao Lu Ruizhe Qian Ning Sun | 2020 | Protein & Cell2020,11,9: | 17 |
| 17 | The chromosome-scale genome reveals the evolution and diversification after the recent tetraploidization event in tea plant显示文摘Tea is one of the most popular nonalcoholic beverages due to its characteristic secondary metabolites with numerous health benefits.Although two draft genomes of tea plant(Camellia sinensis)have been published recently,the lack of chromosome-scale assembly hampers the understanding of the fundamental genomic architecture of tea plant and potential improvement.Here,we performed a genome-wide chromosome conformation capture technique(Hi-C)to obtain a chromosome-scale assembly based on the draft genome of C.sinensis var.sinensis and successfully ordered 2984.7 Mb(94.7%)scaffolds into 15 chromosomes.The scaffold N50 of the improved genome was 218.1 Mb,~157-fold higher than that of the draft genome.Collinearity comparison of genome sequences and two genetic maps validated the high contiguity and accuracy of the chromosome-scale assembly.We clarified that only one Camellia recent tetraploidization event(CRT,58.9–61.7 million years ago(Mya))occurred after the core-eudicot common hexaploidization event(146.6–152.7 Mya).Meanwhile,9243 genes(28.6%)occurred in tandem duplication,and most of these expanded after the CRT event.These gene duplicates increased functionally divergent genes that play important roles in tea-specific biosynthesis or stress response.Sixty-four catechin-and caffeine-related quantitative trait loci(QTLs)were anchored to chromosome assembly.Of these,two catechin-related QTL hotspots were derived from the CRT event,which illustrated that polyploidy has played a dramatic role in the diversification of tea germplasms.The availability of a chromosome-scale genome of tea plant holds great promise for the understanding of genome evolution and the discovery of novel genes contributing to agronomically beneficial traits in future breeding programs. | Jie-Dan Chen Chao Zheng Jian-Qiang Ma Chen-Kai Jiang Sezai Ercisli Ming-Zhe Yao Liang Chen | 2020 | Horticulture Research2020,7,1: | 17 |
| 18 | Comparison of Stretta procedure and toupet fundoplication for gastroesophageal reflux disease-related extra-esophageal symptoms显示文摘AIM: To compare the outcomes between the Stretta procedure and laparoscopic toupet fundoplication(LTF) in patients with gastroesophageal reflux disease(GERD)-related extra-esophageal symptoms.METHODS: From January 2011 to February 2012, a total of 98 patients diagnosed with GERD-related extra-esophageal symptoms who met the inclusion criteria were enrolled in this study. All patients who either underwent the Stretta procedure or LTF treatment have now completed the 3-year follow-up. Primary outcome measures, including frequency and severity of extra-esophageal symptoms, proton pump inhibitor(PPI) use, satisfaction, and postoperative complications, were assessed. The results of the Stretta procedure and LTF therapy were analyzed and compared.RESULTS: There were 47 patients in the Stretta group and 51 patients in the LTF group. Ninety patients were available at the 3-year follow-up. The total of the frequency and severity scores for every symptom improved in both groups(P < 0.05). Improvementin symptom scores of cough, sputum, and wheezing did not achieve statistical significance between the two groups(P > 0.05). However, the score for globus hysterics was different between the Stretta group and the LTF group(4.9 ± 2.24 vs 3.2 ± 2.63, P < 0.05). After the Stretta procedure and LTF treatment, 29 and 33 patients in each group achieved PPI therapy independence(61.7% vs 64.7%, P = 0.835). The patients in the LTF group were more satisfied with their quality of life than those in the Stretta procedure group(P < 0.05). | 19 | Sedimentary Characteristics and Paleoenvironment of Shale in the Wufeng-Longmaxi Formation, North Guizhou Province, and Its Shale Gas Potential显示文摘The Paleozoic Wufeng-Longmaxi shale is one of the main horizons for shale gas exploration in Sichuan Basin. Outcrop, core and thin section observations, X-ray diffraction analysis, trace element geochemistry and other methods have been used to understand the sedimentary characteristics and identify hydrocarbon source rocks in suitable sedimentary paleoenvironments in the Wufeng-Longmaxi shale in northern Guizhou Province. The thickness of the Wufeng-Longmaxi Formation ranges from 20 to 200 m and it was mainly deposited on a deep-water shelf. The TOC content is high, up to 5.75%. The main non-organic minerals are detrital quartz and clay minerals, with a little plagioclase feldspar, potassium feldspar, calcite, dolomite and pyrite. There is also biogenic microcrystalline quartz. Six lithofacies have been identified: siliceous shale, clay shale, calcareous shale, silty shale, carbonaceous shale, and muddy siltstone. Using biological Ba, V/(V+Ni), TOC, V/Cr, B, Sr/Ba and other indicators, we estimate primary productivity, redox conditions and paleosalinity and show that the early stage of Wufeng-Longmaxi deposition occurred under strong anoxic conditions, high paleosalinity and yielded a high TOC content and an excellent potential shale gas source. The anoxic environment was destroyed at the late stages of Wufeng-Longmaxi deposition, the TOC content decreased, so that it is likely to be a high quality source rock. Organic pores acted as the key reservoir space in the shales, and the pores are mainly mesopose, with most pore diameters less than 20 nm.
