| 2 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 6 | Yield and Nicotine Content of Flue-Cured Tobacco as Affected by Soil Nitrogen Mineralization显示文摘Nitrogen(N)supply is the most important factor affecting yield and quality of flue-cured tobacco(FCT).A field experiment and an in situ incubation method were used to study the effects of soil N mineralization in the later stages of growth on yield and nicotine content of FCT in Fenggang and Jinsha,Guizhou Province.The yield and market value of FCT at Fenggang were much lower than those at Jinsha.However,the nicotine content of middle and upper leaves was much higher at Fenggang than at Jinsha when the same rate of fertilizer N was applied,which might be due to a higher N supply capacity at the Fenggang site.At later stages of growth(7-16 weeks after transplanting),the soil net N mineralization at Fenggang(56 kg N ha^(-1))was almost double that at Jinsha(30 kg N ha^(-1)).While soil NH_4-N and NO_3-N were almost exhausted by the plants or leached 5 weeks after transplanting,the N taken up at the later growth stages at Fenggang were mainly derived from soil N mineralization,which contributed to a high nicotine content in the upper leaves.The order of soil N contribution to N buildup in different leaves was:upper leaves>middle leaves>lower leaves.Thus,soil N mineralization at late growth stages was an important factor affecting N accumulation and therefore the nicotine content in the upper leaves. | JU Xiao-Tang CHAO Feng-Chun LI Chun-Jian JIANG Rong-Feng P. Those with igneous characteristics yielded U-Pb ages of 280-2800 Ma, recording a long history of magmatic activity in the region. Zircon Hf isotopic compositions show an abrupt change at ~420 Ma, indicating that prior to that time the magmas came from both ancient and juvenile sources, whereas younger magmas were derived mainly from juvenile material. This may imply that the lithosphere was signifi- cantly modified in composition by a rapid addition of melt from the mantle. | 12 | Reversal of hyperglycemia in diabetic rats by portal vein transplantation of islet-like cells generated from bone marrow mesenchymal stem cells显示文摘AIM: To study the capacity of bone marrow mesenchymal stem cells (BM-MSCs) trans-differentiating into islet-like cells and to observe the effect of portal vein transplantation of islet-like cells in the treatment of streptozotocin-induced diabetic rat. METHODS: BM-MSCs were isolated from SD rats and induced to differentiate into islet-like cells under defined conditions. Differentiation was evaluated with electron microscopy, RT-PCR, immunofluorescence and flow cytometry. insulin release after glucose challenge was tested with ELiSA. Then allogeneic islet-like cells were transplanted into diabetic rats via portal vein. Blood glucose levels were monitored and islet hormones were detected in the liver and pancreas of the recipient by immunohistochemistry. RESULTS: BM-MSCs were spheroid adherent monolayers with high CD90, CD29 and very low CD45 expression. Typical islet-like cells clusters were formed after induction. Electron microscopy revealed that secretory granules were densely packed within the cytoplasm of the differentiated cells. The spheroid cells expressed islet related genes and hormones. The insulin-positive cells accounted for 19.8% and mean fluorescence intensity increased by 2.6 fold after induction. The cells secreted a small amount of insulin that was increased 1.5 fold after glucose challenge. After transplantation, islet-like cells could locate in the liver expressing islet hormones and lower the glucose levels of diabetic rats during d 6 to d 20.CONCLUSION: Rat BM-MSCs could be transdifferentiated into islet-like cells in vitro . Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 18 | Cancer stem cell markers correlate with early recurrence and survival in hepatocellular carcinoma显示文摘AIM:To investigate whether expression of cancer stem cell(CSC)markers is associated with recurrence and survival in hepatocellular carcinoma(HCC)patients.METHODS:A consecutive series of 90 HCC patients who underwent curative hepatectomy between April2007 and April 2009 were analyzed.Of the 90 patients,38(42%)experienced recurrence within two years of surgery.To adjust for baseline differences between this early recurrence group and the other patients,propensity-score matching was used to generate 25 pairs of patients.Immunohistochemistry was used to compare expression of CD133,CD90,and epithelial cell adhesion molecule(EpCAM)in liver tissues from propensity score-matched patients and from 10 healthy adults.Associations of the three markers with HCC,clinicopathological characteristics,early recurrence,and survival time were explored.RESULTS:The expression of all three CSC markers was significantly higher in HCC tissue than in healthy liver tissue(P<0.001 for all).Among the HCC clinicopathology characteristics examined,the absence of tumor capsule was associated with CD133 expression(P=0.005);higher histopathology grade and larger tumor size were associated with CD90 expression(P=0.010 and 0.034,respectively);and elevated serum alpha-fetoprotein levels were associated with EpCAM expression(P=0.021).Expression of CD90 and EpCAM was significantly higher in the early recurrence group than in other patients(P=0.001 and 0.045,respectively),whereas CD133 expression was not significantly different between the two groups(P=0.440).Multivariate analysis identified only CD90 expression as significantly associated with early recurrence.Log-rank analysis identified expression of both CD90 and EpCAM as significantly associated with survival time of HCC patients.Cox regression identified EpCAM expression as an independent predictor of survival time.CONCLUSION:Expression of CD133,CD90,and EpCAM CSC markers may be linked to HCC tumor onset and/or progression.In addition,EpCAM expression is associated with shorter survival time,while CD90 expression is associated with early HCC recurrence. | Zhe Guo Le-Qun Li Jing-Hang Jiang Chao Ou Li-Xia Zeng Bang-De Xiang | 2014 | World Journal of Gastroenterology2014,20,8: | 18 |
| 19 | The chromosome-scale genome reveals the evolution and diversification after the recent tetraploidization event in tea plant显示文摘Tea is one of the most popular nonalcoholic beverages due to its characteristic secondary metabolites with numerous health benefits.Although two draft genomes of tea plant(Camellia sinensis)have been published recently,the lack of chromosome-scale assembly hampers the understanding of the fundamental genomic architecture of tea plant and potential improvement.Here,we performed a genome-wide chromosome conformation capture technique(Hi-C)to obtain a chromosome-scale assembly based on the draft genome of C.sinensis var.sinensis and successfully ordered 2984.7 Mb(94.7%)scaffolds into 15 chromosomes.The scaffold N50 of the improved genome was 218.1 Mb,~157-fold higher than that of the draft genome.Collinearity comparison of genome sequences and two genetic maps validated the high contiguity and accuracy of the chromosome-scale assembly.We clarified that only one Camellia recent tetraploidization event(CRT,58.9–61.7 million years ago(Mya))occurred after the core-eudicot common hexaploidization event(146.6–152.7 Mya).Meanwhile,9243 genes(28.6%)occurred in tandem duplication,and most of these expanded after the CRT event.These gene duplicates increased functionally divergent genes that play important roles in tea-specific biosynthesis or stress response.Sixty-four catechin-and caffeine-related quantitative trait loci(QTLs)were anchored to chromosome assembly.Of these,two catechin-related QTL hotspots were derived from the CRT event,which illustrated that polyploidy has played a dramatic role in the diversification of tea germplasms.The availability of a chromosome-scale genome of tea plant holds great promise for the understanding of genome evolution and the discovery of novel genes contributing to agronomically beneficial traits in future breeding programs. | Jie-Dan Chen Chao Zheng Jian-Qiang Ma Chen-Kai Jiang Sezai Ercisli Ming-Zhe Yao Liang Chen | 2020 | Horticulture Research2020,7,1: | 17 |
| 20 | Secondary Hardening, Austenite Grain Coarsening and Surface Decarburization Phenomenon in Nb-Bearing Spring Steel显示文摘The secondary hardening, the austenite grain coarsening and the surface decarburization phenomenon of Nb-bearing spring steel were investigated, and the effects of niobium on tempered microstructure was studied using scanning electron microscope. The results show that the micro-addition of niobium increases the tempering resistance and produces secondary hardening. The effect of niobium on the size and distribution of cementite particles is one of the primary reasons to increase the hardness after tempering. The grain-coarsening temperature of the spring steel is raised 150 ℃ due to Nb-addition. Furthermore, both the secondary hardening and the austenite grain coarsening phenomenon congruously demonstrate niobium begins observably dissolving above 1 100 ℃ in the spring steel.
