2Helicobacter pylori and oral pathology:Relationship with the gastric infection显示文摘Helicobacter pylori(H.pylori)has been found in the oral cavity and stomach,and its infection is one of the most frequent worldwide.We reviewed the literature and conducted a Topic Highlight,which identified studies reporting an association between H.pylori-infection in the oral cavity and H.pylori-positive stomach bacterium.This work was designed to determine whether H.pylori is the etiologic agent in periodontal disease,recurrent aphthous stomatitis(RAS),squamous cell carcinoma,burning and halitosis.Record selection focused on the highest quality studies and meta-analyses.We selected 48 articles reporting on the association between saliva and plaque and H.pylori-infection.In order to assess periodontal disease data,we included 12 clinical trials and 1 meta-analysis.We evaluated 13 published articles that addressed the potential association with RAS,and 6 with squamous cell carcinoma.Fourteen publications focused on our questions on burning and halitosis.There is a close relation between H.pylori infection in the oral cavity and the stomach.The mouth is the first extra-gastric reservoir.Regarding the role of H.pylori in the etiology of squamous cell carcinoma,no evidence is still available.Isabel Adler Andrea Muio Silvia Aguas Laura Harada Mariana Diaz Adriana Lence Mario Labbrozzi Juan Manuel Muio Boris Elsner Alejandra Avagnina Valeria Denninghoff 2014World Journal of Gastroenterology2014,20,29:21
3Mechanisms of regulation of PFKFB expression in pancreatic and gastric cancer cells显示文摘Enzymes 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase-3 and-4(PFKFB-3 and PFKFB-4)play a significant role in the regulation of glycolysis in cancer cells as well as its proliferation and survival.The expression of these mRNAs is increased in malignant tumors and strongly induced in different cancer cell lines by hypoxia inducible factor(HIF)through active HIF binding sites in promoter region of PFKFB-4 and PFKFB-3 genes.Moreover,the expression and hypoxia responsibility of PFKFB-4 and PFKFB-3 was also shown for pancreatic(Panc1,PSN-1,and MIA PaCa-2)as well as gastric(MKN45 and NUGC3)cancer cells.At the same time,their basal expression level and hypoxia responsiveness vary in the different cells studied:the highest level of PFKFB-4 protein expression was found in NUGC3 gastric cancer cell line and lowest in Panc1 cells,with a stronger response to hypoxia in the pancreatic cancer cell line.Overexpression of different PFKFB in pancreatic and gastric cancer cells under hypoxic condition is correlated with enhanced expression of vascular endothelial growth factor(VEGF)and Glut1 mRNA as well as with increased level of HIF-1αprotein.Increased expression of different PFKFB genes was also demonstrated in gastric,lung,breast,and colon cancers as compared to corresponding nonmalignant tissue counterparts from the same patients,being more robust in the breast and lung tumors.Moreover,induction of PFKFB-4 mRNA expression in the breast and lung cancers is stronger than PFKFB-3mRNA.The levels of both PFKFB-4 and PFKFB-3 proteins in non-malignant gastric and colon tissues were more pronounced than in the non-malignant breast and lung tissues.It is interesting to note that Panc1and PSN-1 cells transfected with dominant/negative PFKFB-3(dnPFKFB-3)showed a lower level of endogenous PFKFB-3,PFKFB-4,and VEGF mRNA expressions as well as a decreased proliferation rate of these cells.Moreover,a similar effect had dnPFKFB-4.In conclusion,there is strong evidence that PFKFB-4 and PFKFB-3 isoenzymes are induced under hypoxia in pancreatic and other cancer cell lines,are overexpressed in gastric,colon,lung,and breast malignant tumors and undergo changes in their metabolism that contribute to the proliferation and survival of cancer cells.Thus,targeting these PFKFB may therefore present new therapeutic opportunities.Oleksandr H Minchenko Katsuya Tsuchihara Dmytro O Minchenko Andreas Bikfalvi Hiroyasu Esumi 2014World Journal of Gastroenterology2014,20,38:17

8Developing a donation after cardiac death risk index for adult and pediatric liver transplantation显示文摘AIM To identify objective predictive factors for donor after cardiac death(DCD) graft loss and using those factors, develop a donor recipient stratification risk predictive model that could be used to calculate a DCD risk index(DCD-RI) to help in prospective decision making on organ use.METHODS The model included objective data from a single institute DCD database(2005-2013, n = 261). Univariate survival analysis was followed by adjusted Cox-regressional hazard model. Covariates selected via univariate regression were added to the model via forward selection, significance level P = 0.3. The warm ischemic threshold was clinically set at 30 min. Points were given to each predictor in proportion to their hazard ratio. Using this model, the DCD-RI was calculated. 10Bifunctional chimeric SuperCD suicide gene -YCD: YUPRT fusion is highly effective in a rat hepatoma model显示文摘AIM: To investigate the effects of catalytically superior gene-directed enzyme prodrug therapy systems on a rat hepatoma model.METHODS: To increase hepatoma cell chemosensitivity for the prodrug 5-fluorocytosine (5-FC), we generated a chimeric bifunctional SuperCD suicide gene, a fusion of the yeast cytosine deaminase (YCD) and the yeast uracil phosphoribosyltransferase (YUPRT) gene.RESULTS: In vitro stably transduced Morris rat hepatoma cells (MH) expressing the bifunctional SuperCD suicide gene (MH SuperCD) showed a clearly marked enhancement in cell killing when incubated with 5-FC as compared with MH ceils stably expressing YCD solely (MH YCD) or the cytosine deaminase gene of bacterial origin(MH BCD), respectively. In vivo, MH SuperCD tumors implanted both subcutaneously as well as orthotopically into the livers of syngeneic ACI rats demonstrated significant tumor regressions (P<0.01) under both high dose as well as low dose systemic 5-FC application,whereas MH tumors without transgene expression (MH naive) showed rapid progression.
